黄芪及其它中草药对双歧杆菌增菌的影响研究

摘要
双歧杆菌是专性厌氧菌,营养条件要求很高,对低pH的抵抗能力较差,生存期较短,这些给工业化生产双歧杆菌发酵制剂带来很大的困难。本课题针对双歧杆菌的这些特点,以长双歧杆菌和两歧双歧杆菌为研究对象,以黄芪为主要材料,较系统地研究了黄芪及其它中药对双歧杆菌生长的影响,以期为进一步开发利用中药资源、双歧杆菌的培养及其制品开发提供理论依据。
     本试验选取的中草药都为药食同源或对人体具有保健功能的药品,采用比浊法和Hungate厌氧滚管技术计数从多种中草药中选取了促进双歧杆菌生长的黄芪。通过黄芪在体外对双歧杆菌生长的影响试验表明,黄芪浸提液在体外对长双歧杆菌Blm和两歧双歧杆菌Bbm的生长具有促进作用,其效果随着黄芪浸提液添加量的增加而增加,并且对不同菌种的促生长效果存在差异。研究了以黄芪浸提液为主要原料的双歧杆菌培养基,大量试验结果表明,在黄芪浸提液中添加大豆蛋白胨、胰蛋白胨、牛肉膏、玉米浆和磷酸氢二钾等成分,对双歧杆菌有明显的促进生长作用,利用黄芪浸提液、大豆蛋白胨、玉米浆和牛肉膏设计了四因素三水平的正交试验,确定了黄芪复合增菌培养基的优化配方:12.5%黄芪浸提液,牛肉膏0.6%,玉米浆0.4%,0.5%大豆蛋白胨,0.2%磷酸氢二钾。在黄芪复合增菌培养基中,经37℃恒温厌氧培养16h,Blm活菌数达到4.59×10~9cfu/mL,Bbm活菌数达到2.67×10~9cfu/mL。
     通过一种选择性双歧杆菌培养基,以健康小白鼠为实验对象,研究了黄芪在动物肠道内对双歧杆菌的影响,动物实验结果表明,黄芪浸提液在体内对双歧杆菌的生长有选择性促进作用,而肠道中主要致病菌—大肠杆菌的生长受到抑制,黄芪中的多糖类物质可能对黄芪在体内选择性地促进双歧杆菌生长起了主要作用,此外,黄芪浸提液还具有明显的整肠作用,同对照组相比,饲喂黄芪浸提液的小鼠排便顺利,粪便颗粒大且湿润。
Bifidobacteria are strict anaerobic germs, hard to nutritional condition, not tolerant to low pH, and short survivibility. This resulted in the great difficulty for Bifidobacterium fermenting dairy products in industrialization. aiming at the peculiarity of Bifidobacterium , the tasks studied the effects of Bifidobacterium Longum and Bifidobacterium bifidus's extract on the growth of Bifidobacterium in order to provide theeoritical foundation for the utilization of Astragalus mongholicus Bunge resource ,using Astragalus mongholicus Bunge as experimental material and Bifidobacterium as strain tested.
     This experimental selection's Chinese medicine has the health care function drugs for the medicine food homology or to the human body, used the turbidimetric method and Hungate anaerobic rolls the tube technology counting to select the Astragalus which from many kinds of Chinese medicine a promotion of Bifidobacteria growth. The influence experiment which grows to Bifidobacteria indicated through the Astragalus in vitro that the Astragalus leaching solution has the promoter action in vitro to Bifidobacterium Longum (Blm) and the two things which do not tally Bifidobacterium bifidus (Bbm) growth, its effect increases along with the Astragalus leaching solution recruitment increases, and presses the growth effect existence difference to the different mold mushroom spawn. Has studied take the Astragalus leaching solution as the primary data Bifidobacterium culture medium, the massive test result indicated that increases ingredients and so on soy protein peptone, pancreas protein peptone, cow meat extract, corn thick liquid and dipotassium hydrogen phosphate in the Astragalus leaching solution, has the obvious promotion growth function to Bifidobacterium, using the Astragalus leaching solution, the soy protein peptone, the corn thick liquid and the cow meat extract has designed four factor three level orthogonal tests, had determined the Astragalus compound increases the fungus culture medium the optimized formula: 12.5% radix astragali leaching solutions, beef extract0.6%, corn thick liquid 0.4%, maize hydrolyzates 0.5%, K_2HPO_40.2%. and the live counts cultivated in the optimum enrichment medium for 16h at 37℃were 4.59×10~9 cfu/mL of Blm and 2.91×10~9 cfu/mL of Bbm.
     Through one selective Bifidobacterium culture medium, take the healthy white mouse as the experimental subject, studied the radix astragali in the animal intestinal tract to Bifidobacterium 's influence, the animal experimentation result had indicated that the radix astragali leaching solution had the selectivity promoter action in vivo to Bifidobacterium 's growth, but in the intestinal tract the main pathogenic bacteria backwoods coli's growth received suppresses, in the radix astragali multi-carbohydrate material possibly grew to the radix astragali at in vivo selectively promotion Bifidobacterium the leading role, in addition, the radix astragali leaching solution also had the obvious entire intestines function, compared with the control group, feeds the radix astragali leaching solution the mouse bowel movement to be smooth, the excrement pellet was big and is moist.
