蜂王浆中活性小分子物质2,6-二叔丁基苯酚的抗衰老作用研究

摘要
蜂王浆中含有多种活性物质,成份比较复杂。蜂王浆是一种传统的营养品及药品,能提高机体的免疫力,抗氧化及延缓机体衰老等。随着年龄的增长,人体会产生较多的自由基,而自由基是致使机体衰老的主要原因之一。同时,自由基也会引起基因功能的改变从而引起多种疾病,如致癌物必须转变成自由基才能起作用。本文为了深入探讨蜂王浆的抗衰老功能因子,分别采用邻苯三酚法和Fenton法测定了蜂王浆中存在的2,6-二叔丁基苯酚对超氧阴离子和羟自由基的清除率;利用高效液相色谱测定了蜂王浆中2,6-二叔丁基苯酚的含量;又通过动物模型实验,测定了2,6-二叔丁基苯酚对小鼠血中总超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活性的影响及对肝脏中丙二醛(MDA)含量的影响。各实验结果如下:
     通过对蜂王浆中2,6-二叔丁基苯酚的高效液相色谱的定量分析,结果显示蜂王浆中的2,6-二叔丁基苯酚含量是12ug/kg。
     2,6-二叔丁基苯酚对超氧自由基和羟自由基有良好的清除作用。当2,6-二叔丁基苯酚的浓度达到600mg/L时,对超氧阴离子的清除率达75.86%,为最强;当2,6-二叔丁基苯酚的浓度达到500mg/L时,对羟自由基的清除率达到84.47%,为最强。
     与空白对照组小鼠比较,饲喂2,6-二叔丁基苯酚、低、中剂量组(蜂王浆+2,6-二叔丁基苯酚)小鼠肝脏中丙二醛含量显著下降。
     与空白对照组小鼠比较,饲喂2,6-二叔丁基苯酚、蜂王浆组、中剂量组(蜂王浆+2,6-二叔丁基苯酚)小鼠血中SOD的含量及活性显著上升。
     与空白对照组小鼠比较,饲喂2,6-二叔丁基苯酚、低、中、高剂量组(蜂王浆+2,6-二叔丁基苯酚)小鼠血中GSH-PX的含量及活性上升。
The royal jelly includes the many kinds of active material, the ingredient is quite complex. The bee royal jelly is one traditional nutriment and the drugs, can enhance the organism the immunity, anti- oxidizes and delays theorganism to be senile and so on.Along with the age growth, the person produces more free radicals, the free radical causes one of organism senile main reasons, simultaneously, the free radical also can cause the gene function change, thus causes the many kinds of diseases,as the careinogen must be transformed the free radical in order to play the role. This article in order to thoroughly discusses the bee royal jelly the antisenile function factor, separately used the pyrogallic acid method and the Fenton method that has determined Butylated Hydroxytoluene which in the bee royal jellyexisted, to ultraoxygen and hydroxy free radical elimination percentage; Used the highly effective liquid chromatography to determine in the royal jelly the Butylated Hydroxytoluene content; Also through in vitro animal modelexperiment, has determined Butylated Hydroxytoluene to the mouse blood in total SOD, the GSH peroxide enzyme activity influence and to the liver of the MDA content influence. Its each experimental result as follows:
     Through to the Butylated Hydroxytoluene in the bee royal jelly with highly effective liquidchromatography quantitative analysis, the result demonstrates that royal jelly Butylated Hydroxytoluene content is 12ug/kg.
     Butylated Hydroxytoluene has the good scavenging action to the ultra oxygen free radicaland the hydroxy free radical. When the Butylated Hydroxytoluene density achieved to 600mg/L, elimination percentage reaches 75.86% to the ultraoxygen, which is strongest; When the Butylated Hydroxytoluene density achieved to 500mg/L, elimination percentage achieves 84.47% to the hydroxy free radical, which is strongest.
     Comparing with the blank comparison group mouse, Butylated Hydroxytoluene, low, middle in the monitoring group liver of the MDA content are remarkable drop.
     Comparing with the blank comparison group mouse, Butylated Hydroxytoluene, royal jelly, middle in monitoring group, blood SOD content and active are remarkable rise.
     Comparing with the blank comparison group mouse, Butylated Hydroxytoluene, low, middle, high group, GSH-PX content and the active are rise.
