希美宁对2型糖尿病高脂血症模型大鼠血脂、血流变及血清Leptin的影响

摘要
目的:研究希美宁对2型糖尿病(diabetes mellitus,DM)合并高脂血症(hyperlipidemia,HLP)大鼠的空腹血糖(fasting blood glucose,FBG)、血清胰岛素(insulin,INS)、糖化血红蛋白(glyxosylated hemoglobi,HbAIC)、血脂、血流变及血清瘦素(leptin)的影响,并探讨其作用机制。
     方法:将80只SD雄性大鼠随机分为两组,即空白组和糖尿病组。空白组以普通饲料喂养,糖尿病组以高糖高脂饲料喂养。8周后,糖尿病组大鼠一次性腹腔注射小剂量链脲佐菌素(streptozotozin,STZ),损坏部分胰腺,诱发高血糖。1周后测血糖,以空腹血糖>16.7mmol/L的大鼠为成功模型纳入实验。将纳入实验的糖尿病高脂血症大鼠随机分为糖尿病模型组、希美宁组、血脂康组。希美宁组和血脂康组按体重不同以成人剂量的0.018倍相应灌胃;空白组和模型组以等体积生理盐水给大鼠灌胃。14周末给药结束后,隔夜空腹摘眼球取血,收集血液,分装不同试管,测定血糖、血流变的水平,分离血清,以测定糖化血红蛋白、胰岛素、血脂及Leptin的水平。
     结果:①采用高糖高脂饮食诱导+STZ综合造模法的模型组大鼠其血糖、血清胰岛素、糖化血红蛋白、血脂、血流变、Leptin水平明显高于空白组,有统计学意义;②血脂康组治疗后的大鼠,其血糖、血清胰岛素、糖化血红蛋白、血脂、血流变、Leptin水平较模型组显著降低,有统计学意义;③希美宁组其血糖、血清胰岛素、糖化血红蛋白、血脂、血流变、Leptin水平低于血脂康组,有统计学意义。
     结论:采用高糖高脂饮食诱导+STZ综合造模法成功复制糖尿病合并高脂血症大鼠的模型;希美宁对糖尿病合并高脂血症大鼠的血糖、血清胰岛素、糖化血红蛋白、血脂、血流变、Leptin有显著治疗作用,其作用机制之一可能是通过抑制Leptin过度表达实现的。
Objective.To study the effects of Ximeining on blood lipid、hemorrheology and Leptin in serum of 2DM mice with hyperlipoidemia,and investigate its mechanism.
     Methods:80 male rats were randomly divided into two groups,the blank group and diabetes group.To the control group fed normal diet,diabetes group high sugar and high fat diet.After eight weeks,the diabetes group was injected with low-dose streptozotocin(STZ),damaged part of the pancreas,induced by high blood sugar.One week later measure blood glucose,fasting plasma glucose(FBG)>16.7mmol/L,which were choosed into the model for successful experiments.Experimental diabetes will be divided into the model of diabetic rats,the XIMEINING group,and the XUEZHIKANG group.With saline gavage to blank and model group mice,the XIMEINING group and the XUEZHIKANG group treatment by different weight to adult dose of 0.018 times the corresponding administration.After 14 weekends administration,kill the animals by picking the overnight fasting blood eyeball,collecting the blood with different sub-tube,separating the serum,to determinate the level of glucose, glycated hemoglobin,serum insulin,blood lipids,hemorrheolog and Leptin.
     Results:With high sugar and high fat diet + STZ integrated model of the mice model of their blood sugar,blood lipid,hemorrheolog and Leptin.Levels were significantly higher than the control group.With the ximeining group their blood sugar,blood lipid,hemorrheolog and TNF-αlevel compered with the model were significantly lower,Blood sugar and Leptin levels of XIMEINING group were lower than xuezhikang treatment group.It was statistically significant,blood lipid levels of XIMEINING group decreased compared with XUEZHIKANG group.It was statistically significant.
     Conclusion:High sugar and high fat diet + STZ integrated model of successful replication of diabetic hyperlipidemia in the modeI,XIMEINING lipid metabolism of diabetic mice disorder of the blood sugar,blood lipid, hemorrheolog and Leptin treatment of a significant role.It's role is imposible one of the mechanisms by inhibiting Leptin to achieve the over-expression.
