紫石英对排卵障碍大鼠模型卵巢局部INH、IGF-Ⅰ影响机制的研究

摘要
目的:探讨中药紫石英对排卵障碍大鼠模型卵巢局部INH、IGF-I影响的作用机制以及紫石英药理作用的物质基础,为紫石英在临床上治疗排卵障碍性不孕提供理论依据。
     方法:选用未成年24d龄SD雌性大鼠60只,体重75±5g,按随机数字表法分为模型组、克罗米芬组、补肾活血方组、紫石英组、正常组、氟化钙组,每组10只。用Bogovich法建立排卵障碍大鼠病理模型。以补肾活血方及克罗米芬(CC)为对照,用免疫组化法观察紫石英对排卵障碍大鼠模型卵巢局部抑制素(INH)、胰岛素样生长因子-I(IGF-I)蛋白表达的影响;并将紫石英组与氟化钙组的实验结果进行比较以分析紫石英产生相应药理作用的可能物质基础。
     结果:模型组与正常组比较,卵巢局部组织中IGF-I与INH在表达强度上呈异常高表达,差异具有显著性(P<0.01)。经过紫石英、补肾活血方、克罗米芬及氟化钙治疗后,各组卵巢局部组织中IGF-I与INH的蛋白表达均明显降低(P<0.01,P<0.05),说明紫石英、补肾活血方、克罗米芬及氟化钙对IGF-I与INH的表达都具有抑制作用。但紫石英组、补肾活血方组及克罗米芬组两两比较,差异无统计学意义(P>0.05)。紫石英组大鼠卵巢局部IGF-I与INH平均光密度值虽均较氟化钙组要小,但两组比较差异无统计学意义。
     结论:IGF-I与INH可能参与了排卵障碍的发病机制。紫石英能调节排卵障碍大鼠卵巢局部IGF-I与INH的异常高表达且效果稍优于氟化钙。提示对调节卵巢局部IGF-I与INH的异常高表达,紫石英要较单纯氟化钙作用强;紫石英可能具有内分泌激素样作用,能够使下丘脑-垂体-卵巢轴(HPOA)的调节功能得以改善,从而促进初级卵泡向优势卵泡发育,但其产生相应作用的物质基础有待进一步研究。
Objective:This paper explores the material basis of the fluoritum's influence of the rat models with ovulation failure on ovulation inhibition(INH), insulin-like growth factor-I(IGF-I) and ovarian morphology. It provides the theoretical foundation for clinical treatment of ovulation failure.
     Methods:60 SD female rats of 24 days age were chose as research object, and were distributed into 6 groups at random, which were model control group, western medicine control group, fluoritum group, calcium fluoride group, norishing kidney prescription, and normal control group, each group had ten rats. Using the Bogovich method set a pathological rat model of ovulation failure, using the Clomiphene and kidney tonify and blood circulation simulating soup for contracting, and using the immunohistochemicalstaining observed the fluoritum's influence on the rat with ovarian failure part of the INH and IGF-I.
     Result:Compared with the normal group, the model group's intensity of expression of IGF-I and INH is higher. The difference is obvious (P <0.01). After the treatment, the intensity of expression of IGF-I and INH reduced obviously(P<0.01, P<0.05). It shows that the fluoritum,calcium fluoride, norishing kidney prescription and medicine can repress the intensity of expression of IGF-I and INH. the above four groups have no difference (P>0.05). This illustrates calcium fluoride and other microelement jointly take effect in reducing the intensity of expression of IGF-I and INH. The average of fluoritum rats ray density is obviously smaller than calcium fluoride rats, but it's not a significant difference.
     Conclusion:The above result of the experiment indicates that IGF-I and INH probably participate the nosogenesis of polycystic ovary syndrome (PCOS). Fluoritum can reduce the abnormal high expression in local ovary of ovulation obstruction rat, and attenuate the function of the IGF-I and INH in local ovary, and it is superior to CaF2 in the intensity. CaF2 and the other elements of fluoritum maybe restrain the level of IGF-I and INH. It shows that fluoritum probably has the function of endocrine hormone. It can improve the function of regulate of HPOA, and promote the growth of primary ovarian follicles to dominant follicle, but the foundation material of fluoritum needs further study.
