绵羊BMP15和BMPR-IB基因多态性研究

摘要
本研究以骨形态发生蛋白15(BMP15)和骨形态发生蛋白受体(BMPR-IB)为候选基因,以内蒙古地方品种蒙古羊、内蒙古细毛羊、呼伦贝尔羊、中国美利奴羊(科尔沁型)为研究对象,对在生殖中起重要作用的BMP15基因和BMPR-IB基因进行多态性检测,以比较它们在不同绵羊品种中的多态性,旨在寻找与繁殖力相关的遗传标记,为内蒙古绵羊选育和改良提供理论依据。
     (1)通过对GenBank中绵羊BMP15基因作比对分析,在其外显子1序列内设计一对引物,采用PCR技术扩增其部分序列并进行PCR-SSCP多态性分析。实验结果在4个绵羊品种中均检测到BMP15的突变,且检测到的BMP15的突变在4个绵羊品种中均呈现三种基因型。分析实验结果显示:BMP15基因外显子1中虽然存在突变位点,但这一突变位点对绵羊繁殖力高低没有显著影响。
     (2)利用GenBank中绵羊的BMPR-IB序列信息,通过序列比对、分析,采用PCR技术扩增其部分序列并进行PCR-RFLP多态性分析。实验结果表明:繁殖力较高的中国美利奴羊(科尔沁型)BMPR-IB基因编码序列第746位碱基处发生了与Booroola Merino绵羊相同的突变(A746G),而繁殖力较低的蒙古羊、呼伦贝尔羊、内蒙古细毛羊、则没有发生这种突变。
Abstract BMP15 and BMPR-IB as candidate genes on the fecundity,The polymorphism of BMP15 gene and BMPR-IB gene were detected in four sheep breeds mongolian sheep、hu lun bei er sheep、Inner mongolian fine wool sheep and china merino( ke er qin type). in order to find a molecular genetic marker which has significance in mongolian sheep breeding.
     (1) the primer was designed by BMP15 exon1 sequence in the GenBank, adopted PCR technique to magnify the sequence and analysis the polymorphism by PCR-SSCP of BMP15 gene in different four breeds of sheep.The results were as follows:The polymorphisms of PCR-SSCP existed in the four ewe brains,and this polymorphism show three gene types in every one brain.AA、A+、++ type;The mutation detected did not affect significantly the prolificacy of ewes.
     (2) the primer was designed by BMPR-IB sequence in the GenBank, adopted PCR technique to magnify the sequence and analysis the polymorphism by PCR-RFLP of BMPR-IB gene in different breeds of sheep. The results in dicated that there was a same mutation (A746G) of BMPR-IB gene in china Merino as that in Booroola Merino ewes. That Mutation did not exist in mongolian sheep、hu lun bei er sheep、Inner mongolian fine wool sheep.
