吸烟大鼠实验性牙周炎动物模型的建立

摘要
目的:通过观察结扎丝结扎吸烟大鼠上颌第二磨牙牙颈部后牙周组织的变化,探讨建立吸烟大鼠的实验性牙周炎模型。
     方法:采用被动吸烟结合结扎丝结扎法制备吸烟大鼠实验性牙周炎模型。健康Wistar大鼠40只随机分为健康对照组(C)、单纯结扎组(L)、单纯吸烟组(S)、吸烟+结扎组(SL),每组均为10只。C组正常喂养,不做任何干预;L组以双股结扎丝结扎上颌第二磨牙牙颈部;S组每天放入吸烟染毒箱被动吸烟2次,12支/次,30min/次。SL组以双股结扎丝结扎上颌第二磨牙牙颈部,每天与S组同时给予被动吸烟。每周观察牙龈健康状况,当实验组大鼠出现明显的牙龈炎症,且探诊深度明显加深时处死动物,通过大体及X线检查观察并记录上颌第二磨牙牙龈指数(GI)、牙周探诊深度(PD)和牙槽骨丧失值(ABL),通过组织学检查观察牙槽骨变化,并通过酶联免疫标记法测定血清和牙龈组织中IL-6含量。
     结果:第6周时,SL组探诊深度明显增加。L组除PD和血清中IL-6含量,其余指标均高于C,S组。SL组各观察值高于S,L组。S组血清中IL-6含量高于C,L组。第8周时,L组探诊深度明显增加。L组、SL组牙龈指数、牙周探诊深度、牙槽骨丧失值、牙龈中IL-6含量均高于C,S组。而SL、L组GI无明显差别,余各观察值均以SL组高。S组、SL组血清IL-6高于C,L组。
     结论:1、被动吸烟+结扎丝结扎法可成功建立吸烟大鼠实验性牙周炎模型。2、第6周时可以形成稳定的吸烟大鼠牙周炎模型。
Subjective:To establish experimental animal model of periodontitis on smoking rats.
     Methods:Forty health Wistar rats were randomly divided into four groups as follow:control group(C),ligation group(L),smoking group(S),ligation and smoking group(SL).Every group was ten.The control group was raised with no treatment.The ligation group was ligated by two erthodontics wires around the cervical of the second molar.The smoking group was subjected to passive smoking box induce to smoke 12 cigarattes every time for half an hour,twice one day.The last group,ligated as the ligation one,smoks together with the smoking one every day.Observe the inflamation of periodontal tissue every week. PD becomes deeper,recording the GI,PB and the bone loss through clinal exam,X-ray and historigical exam.At the same time,IL-6 was determined in the serum and gingival tissue.
     Results:6 weeks later,PD got deeper in SL group.Except PD and the IL-6 content in serum were significantly higher than C,S group.The indexs of SL group were higher. while IL-6 content in serum in S was h.At the 8th week,PD got deeper in L group.every index,except IL-6 content in serum,were higher in L and SL group than their contrast ones.The GI made no difference between the L group and the SL one(p>0.05).IL-6 in serum in the S or SL was still higher(p<0.05).The C group contained.
     Conclusion:1 Both passive smoking and ligation that given on rat can successfully establish animal model of periodontitis.2 Six weeks later,the experience animal model of periodontitis on smoking rats is successful.
