鸡肾型IBV JL株的分离鉴定及免疫原性研究

摘要
鸡传染性支气管炎是由传染性支气管炎病毒(Infectious Bronchitis Virus,IBV)引起的一种急性高度接触性传染病。近几年来,肾型传染性支气管炎在吉林省各地鸡场常有发生,给养禽业生产造成了巨大的损失。为了及时、准确的掌握该疫病发生发展规律,有效地对其进行控制,本研究开展了对吉林地区肾型鸡传染性支气管炎病原的分离鉴定及疫苗研制等工作。
     本试验采集吉林省鸡场疑似肾型传染性支气管炎的发病鸡病料,通过9~11日龄SPF鸡胚进行病毒的分离和传代,并对分离毒株进行了病毒的血凝性、血凝抑制性、致病性、鸡胚矮小化、电镜特征等生物学特性鉴定及理化特性测定。试验结果表明分离株病毒尿囊液经磷酸酯酶处理后均可凝集鸡红细胞,标准阳性血清可特异性的抑制其凝集性,致病力试验可复制出与自然发病相同的病例,病毒传代物有明显的致鸡胚矮小化作用,透射电镜下可见有近似球形、直径100nm左右的冠状病毒粒子,理化特征符合传染性支气管炎特征,且与H120、IBV-X毒株间不能交叉保护。因此判定分离鉴定的IBV毒株为鸡肾型传染性支气管炎病毒,并定名为IBV JL株。
     收获肾型IBV JL株尿囊液经纯化、沉淀等制备病毒抗原,并成功建立了间接ELISA检测方法。选用肾型IBV JL株制备灭活油佐剂疫苗,通过免疫保护试验、免疫效力试验、区域试验和田间扩大试验表明,制备的疫苗免疫效果确实、保护率高,能够有效地预防肾型传染性支气管炎的发生。
Avian Infectious Bronchitis Disease is caused of Infectious Bronchitis Virus(IBV). It is an acute and height contagious disease. Recently, Iinfectious Bronchitis (IB) are epidemic in JiLin province that result in enormous economic loss.
     In order to discover the regularity of genesis and development of this disease, control the epidemic disease, we did some research on isolation and identification and preparation inactive vaccine f Avian Infectious Bronchitis Virus.
     The strains of Infectious Bronchitis Virus(IBV)was isolated from chicken kidney of the young layer flocks in some regions of JiLin province. Biological and physical chemistry characterizations of the virus were investigated by virus pathogenicity to chicken embryo and early chickens, hemagglutination test(HA), hemagglutination inhibition test(HI), dwarfing embryonated chicken eggs and electron microscope.
     It demonstrated that allantoic fluid of each passage virus could hemagglutinate chicken red blood cells by treatment with phosphoesterase. The standard positive serum to IBV could inhibited the hemagglutination. The isolates could make kidney diseases in chickens. The spherical virus covered with spike like corona about 100nm in
