三种内生真菌代谢产物化学成分研究

摘要
本论文包括Penicillium dangeardii Pitt、波兰青霉(Penicillium polonicum K.M.Zalessky)和烟曲霉(Aspergillus fumigatus Fresen.)三株内生真菌次生代谢产物化学成分的研究。
     内生真菌是指一类在其部分或全部生活史中存活于健康植物组织内部,而不使宿主植物表现出明显感染症状的微生物。
     目前,从内生真菌次生代谢产物中已经发现许多的新的化合物具有抗肿瘤、抗菌、抗病毒、杀虫等活性。本课题组曾对仪花(L. rhodostegia)的根——单刀根和格木这两种药用植物的化学成分进行了较系统研究,从中分离得到了间苯三酚、二苯乙烯、黄酮类、咖萨因型二萜酰胺类生物碱类化合物。在此基础上,本课题组对单刀根和格木的内生真菌进行了分离纯化,根据初步的细胞毒活性筛选与化学筛选结果,选择其中发酵物具有活性的菌株Penicillium dangeardii Pitt、波兰青霉(Penicillium polonicum K.M.Zalessky)和烟曲霉(Aspergillus fumigatusFresen.),放大发酵并对其菌液乙酸乙酯部位的化学成分进行研究,从中分离鉴定了37个化合物(其中有3个化合物在不同菌株次生代谢产物中出现),包括4个内酯类化合物,17个二酮哌嗪类化合物,6个甾体类化合物,4个苯甲酸类化合物,3个咖啡酰奎尼酸类化合物,1个腺苷类化合物,1个神经酰胺类化合物和1个单甘油酸酯化合物,通过谱学方法确定其结构分别为:Penicillactone-A(1), Penicillactone-B(2), Penicillactone-C(3), Penicillactone-D(4).3-羟基-苯甲酸-4-O-β-D-吡喃葡萄糖-(1→6)-β-D-呋喃核糖苷(5),8-malonic acid-7-α-D-ribbofuran osyladenine(6),邻苯二甲酸二正丁酯(7),邻苯二甲酸二异丁酯(8),3-咖啡酰基奎尼酸(9),4-咖啡酰基奎尼酸(10),5-咖啡酰基奎尼酸(11),3-羟基-苯甲酸-4-O-β-D-吡喃葡萄糖苷(12),麦角甾醇(13),过氧化麦角甾醇(14),麦角甾-7,22(E)-二烯-3β,5α,6β三醇(15),2,3-二羟基丙基十八酸酯(16),N-(1'-羟甲基-2',3-二羟基)十七烷基-2-羟基二十四碳酰胺(17)8,20-doil-6-MethoxspirotryprostatinB(18), Cyclo-(S-Val-R-Leu)(19), Cyclo-(S-Pro-R-Val)(20), Cyclo-(S-Pro-R-Ile)(21), Cyclo-(S-Pro-R-Leu)(22), Spiro tryprostain A(23); Tryprostatin B(24), fumitremorgin C(25), demethoxyfumitremorgin C(26), Cyclo-(R-Pro-R-Phe)(27),12R,13S-dihydroxyfumit remorginC(28), Cyclo(trans-4-OH-D-Pro-D-Phe)(29), Cyclo-(R-Tyr-S-Ile)(30), Cyclo-(R-Phe-S-Ile)(31), Cyclo-(R-Val-S-Val)(32), Cyclo-(R-Ala-S-Val)(33), Cyclo-(R-Val-S-Tyr)(34);化合物1,2,3,4,5,6,18和19为新化合物,其中化合物1,2,3,4为新骨架类化合物(中文可命名为:青霉内酯A-D)。新结构的单体化合物进行细胞毒活性,抗病毒活性和抗炎活性的筛选。发现化合物1.2具有抗炎活性。
The thesis included three parts of studies on the chemical constituents from the endophytes of Penicillium dangeardii Pitt, Penicillium polonicum K.M.Zalessky and Aspergillus fumigatus Fresen.
     The term'endophyte' is an all-encompassing topographical term which includes all organisms that, during a variable period of their life, symptomlessly colonise the living internal tissues of their hosts. This definition is broad enough to include virtually any organism residing inside a plant host.The compounds from endophyte were bioactivity of antitumor,antibiosis,antivirus and anthemintic.
     The genus Lysidice consists of two species, L. rhodostegia Hance and L. brevicalyx Wei. The chemical constituents from roots of L. rhodostegia has been studied previously, leading to the isolation of some phloroglucinols, stilbenes and flavanoids. Meanwhile, a series of endophytes were isolated from the roots of L. rhodostegia. According to the cytotoxic activity and chemical screening results, the bioactive strain Pencillium dangeardii Pitt, Penicillium polonicum K.M.Zalessky and Aspergillus fumigatus Fresen were selected for scale-up fermentation, and the EtOAc extract of its broth was then chemically investigated, leading to the isolation of34compounds including4lactones,17diketopiperazines,4benzoic acids,3sterols,3caffeoylquinates,1adenosine,1ceramide and1monoglyceride.Though spect roscopic methods,their structures were respectively established as:Penicillactone-A(1), Penicillactone-B(2), Penicillactone-C(3), Penicillactone-D(4),3-hydroxy-benzoic acid-4-O-β-D-glucopyranosyl-(1→6)-β-D-ribbofuranoside(5),8-malonic acid-7-α-D-ribbofuranosyladenine(6), dipropylphthalate(7), diisobutylphthalate(8),3-dicaffeoyl quinate(9),4-dicaffeoylquinate(10),5-dicaffeoy lquinate(11),3-hydroxy-benzoic acid-4-O-β-D-glucopyranoside(12), ergosterol(13), peroxy-ergosterol(14), Ergosta-7,22-trien-3β,5a,6β-triol(15),2,3-dihydroxypropylstearate(16),2-hydroxy-N-(1,3,4-trihydroxydocosan-2-yl)icosanamide(17),8,20-doil-6-MethoxspirotryprostatinB(18), Cyclo-(S-Val-R-Leu)(19), Cyclo-(S-Pro-R-Val)(20), Cyclo-(S-Pro-R-Ile)(21), Cyclo-(S-Pro-R-Leu)(22), Spirotryprostain A(23), Tryprostatin B(24), fumitremorgin C(25), demethoxyfumitremorgin C(26), Cyclo-(R-Pro-R-Phe)(27),12R,13S-dihydroxyfumi tremorginC(28), Cyclo(trans-4-OH-D-Pro-D-Phe)(29), Cyclo-(R-Tyr-S-Ile)(30), Cyc lo-(R-pHE-S-Ile)(31),Cyclo-(R-Val-S-Val)(32), Cyclo-(R-Ala-S-Val)(33), Cyclo-(R- Val-S-Tyr)(34). There are8new compounds(1,2,3,4,5,6,18and19).The compounds of1,2,3and4are the novel skeleton. The novel compounds was assayed for cytotoxic activities, antiviral activities and antiinflammatory.
