千佛菌对环磷酰胺模型小鼠Th1、Th2类细胞因子表达的调节作用

摘要
目的:
     通过研究千佛菌对环磷酰胺免疫抑制模型小鼠的Th1、Th2类细胞因子(IL-2、IL-10) mRNA表达的影响以及外周血白细胞数、胸腺指数、脾脏指数的变化,探讨千佛菌对机体免疫功能的调节作用。
     方法:
     将小鼠随机分为四组,即正常对照组、环磷酰胺模型对照组、千佛菌小剂量组和千佛菌大剂量组,除正常对照组外,其他各组均隔日腹腔注射环磷酰胺,以制备实验模型。复制模型后,按0.4ml/只的剂量对正常对照组和模型对照组用生理盐水灌胃;千佛菌大剂量组和小剂量组分别以100%和50%的药液浓度按照0.4ml/只用千佛菌药液灌胃。14天后,用荧光定量PCR法分别检测小鼠脾脏淋巴细胞的IL-2mRNA、IL-10mRNA的表达水平,并计算小鼠胸腺指数、脾脏指数,以及外周血白细胞数,分别用单因素方差分析法处理各组数据。
     结果:
     环磷酰胺模型组小鼠的IL-2mRNA和IL-10mRNA表达水平、外周血白细胞数、脾脏指数以及胸腺指数,与正常对照组对比较,各项指标均明显较低,两组比较有显著性差异(P<0.01或P<0.05);千佛菌大剂量组与模型组比较以上指标水平表现为明显升高(P<0.01或P<0.05);与模型组比较,千佛菌小剂量组IL-2mRNA、IL-10mRNA表达水平升高,有显著差异(P<0.05),而外周血白细胞数有升高趋势,但与模型组比较无显著性差异(P>0.05);千佛菌大、小剂量组间IL-2mRNA表达水平、脾脏指数、外周血白细胞数比较亦有显著差异(P<0.01或P<0.05)。
     结论:
     千佛菌能提高环磷酰胺免疫抑制小鼠的Th1、Th2型细胞因子(IL-2、IL-10)mRNA的表达水平。
Objective: In order to investigate the immunoregulation of Grifola frondosa, we studied the changes of cyclophosphamide model mice Th1,Th2 type cytokines (IL-2mRNA and IL-10mRNA’s expression).
     Methods: Kunming strain mice were randomly made up with 4 groups: normal control, model group, model treated with high dosage of Grifola frondosa , model treated with low dosage of Grifola frondosa. Other groups were treated with abdomen injection with cyclophosphamide every two days except normal control. Whether the model were successful, the group of normal control and the cyclophosphamide model were administered 0.4ml NS per mice everyday; The other groups treated with different dosage of Grifola frondosa ,which were daily administered 0.4ml drug solution of 100% and 50% concentration respectively. 14 days later, the expression of IL-2mRNA and IL-10mRNA were measured by FQ-PCR. Count the thymus index, spleen index and leukocyte of peripheral blood that data processing were determinate by one-way ANOVA method.
     Results: Cyclophosphamide models were developed successfully, The levels of IL-2mRNA and IL-10mRNA’s expression were decreased very much obviously in model group (P<0.01 or P<0.05), compared with that in control group. The levels of the expression on IL-2mRNA and IL-10mRNA were increased very much significantly in high dosage group of Grifola frondosa (P<0.01 or P<0.05), compared with that in model group. The levels of the expression on IL-2mRNA and IL-10mRNA in serum with low dosage of Grifola frondosa were increased obviously (P<0.05) but the levels of leukocyte of peripheral blood count was no marked (P>0.05) in comparison with that in model group. Also, these was conspicuity difference in levels of expression of IL-2mRNA, spleen index and leukocyte of peripheral blood count in serum between high and low dosage group of Grifola frondosa (P<0.01 or P<0.05).
     Conclusion: Grifola frondosa can enhance significantly levels of IL-2mRNA and IL-10mRNA’s expression serum with cyclophosphamide model mice.
