牛磺酸对大鼠深Ⅱ度烫伤后肝功能的影响

摘要
目的研究牛磺酸对大鼠深Ⅱ度烫伤后肝功能的影响;初步探讨牛磺酸影响大鼠深Ⅱ度烫伤后肝功能的机制。
     方法选用40只SD大鼠,体重220~250克,不分雌雄,随机分为两组,实验组、对照组各20只。10%硫化钠脱去大鼠背毛,以戊巴比妥钠30~40mg/kg腹腔注射麻醉大鼠,将其背部浸入100℃沸水中15秒,造成面积为全身体表总面积30%的深Ⅱ度烫伤。致伤后实验组大鼠即刻给予牛磺酸注射液200mg/kg腹腔注射,对照组给予等剂量的生理盐水腹腔注射。分别于其致伤后的1h、6h、12h、24h、48h处死实验组和对照组大鼠,每个时间点每组处死4只,取动脉血和肝组织。光镜观察肝组织细胞结构形态;测量血中谷丙转氨酶(ALT)、谷草转氨酶(AST)的含量,比较两组大鼠不同时间点之间血清ALT、AST的差异,测定两组大鼠不同时间点组织匀浆中丙二醛(MDA)和超氧化物歧化酶(SOD)的含量,并作统计学分析。
     结果所有选入试验的SD大鼠均存活至试验结束。光镜观察可见烫伤后两组大鼠肝细胞均有不同程度的损伤,对照组较实验组严重。对照组伤后肝组织内肝小叶内中央静脉扩张,汇管区内有淋巴细胞浸润;肝窦扩张充血,肝窦内炎细胞浸润;肝细胞空泡变性、胞核浓缩;肝组织出现凝固性坏死。实验组较对照组有明显改善,组织结构基本接近正常。烫伤后不同时间点大鼠血清谷草转氨酶(AST)的水平,和对照组都较实验组明显升高(P<0.01),两组之间差异有统计学意义。血清谷丙转氨酶(ALT)的水平,1h时实验组和对照组之间无统计学差异(P>0.05);其他各时间点对照组AST的含量较实验组明显升高,两组之间比较均有差异(P<0.01),有统计学意义。实验组各时间点丙二醛(MDA)水平低于对照组,两组之间有统计学差异(P<0.01),差异有显著性。实验组烫伤后不同时间点超氧化物歧化酶(SOD)水平均高于对照组(P<0.01),统计学意义。
     结论通过腹腔注射牛磺酸对大鼠深Ⅱ度烫伤后早期肝功能有保护作用。可能是通过抗脂质过氧化、清除氧自由基的机制保护肝脏。
Objective To study the effect of taurine on protecting the liver function ofdeep partial-thickness burn in rats. To explore the mechanism of taurineeffect on the liver function of burn in rats.
     Method Forty Sprague-Dawley (SD) rats were randomly selected, deeppartial-thickness bums of the body surface were inflicted. The rats wererandomly divided into 2 groups. 20 rats in the experiment group, 20 rats inthe control group. Then the experiment group was treated with abdominalinject of taurine 200mg/kg, and the control group was treat with the samedose of 0.9%saline solution. The two groups be treated were injected onlyonce. Then we killed 4 rats when 1h、6h、12h、24h、48h after the burns in eachgroup, to got the liver tissue and sera. Observed the morphous of the livertissue under light microscope at the each time. Measured and analyzedAlanine aminotransferase(ALT)、Aspartate aminotransferase(AST)、MaleicDialdehyde(MDA) and Superoxide Dismutase(SOD).
     Result The result of light microscope show the two groups treated withbum all have injury, the control group was more severe than the experimentgroup. The contents of ALT and AST, MDA of control group were higherthan the experiment group(P<0.05), SOD of the experiment group was the higher than the control group(P<0.05).
     Conclusion In this experiment, the result show that taurine could defendedthe function of liver of deep partial-thickness bum. It may possibly achievedthis effect though anti-oxidation.
