~(60)Co-γ射线对大鼠睾丸间质细胞的损伤及黄芪多糖对该损伤防护作用的研究

摘要
研究背景及目的:
     随着科技水平及医学放射检查和治疗的普及,人们接触射线的越来越多,由此而导致的睾丸生精功能低下的人群逐年增加。60Co-γ射线照射可致睾丸生精细胞、支持细胞及间质细胞损伤。黄芪多糖(APS)是黄芪的主要有效成分,具有抗辐射、抗氧化作用。本实验旨在观察大剂量60Co-γ射线局部照射SD大鼠睾丸后,其病理结构及分子水平的改变并通过黄芪多糖灌胃组观察APS对60Co-γ射线导致的损伤的影响及其机制。
     方法
     雄性SD大鼠30只,随机分成3组:空白对照组,照射对照组,黄芪多糖灌胃组。光镜对比观察各组大鼠睾丸间质细胞的形态学改变;ELISA法检测血清睾酮(T)、黄体生成素(LH)、过氧化氢酶(CAT)的含量;采用黄嘌呤氧化酶法(羟胺法)测定血清SOD活力,半定量RT-PCR检测睾丸间质细胞黄体生成素受体(LHR)的1mRNA水平。
     结果
     大剂量60Co-Y射线局部照射性腺后大鼠睾丸间质细胞形态发生改变,与正常对照组相比,细胞体积缩小,胞质减少,细胞核皱缩,数目减少,其睾丸组织LHR的mRNA水平与正常对照组相比明显降低(p<0.05);其中空白组LHRmRNA的相对含量为1.18±0.15,照射对照组LHRmRNA的相对含量为0.39±0.17。
     黄芪多糖灌胃组与照射对照组相比睾丸组织病理改变较轻,LHR的mRNA水平基本接近正常水平,相对含量为0.83±0.11;血清中SOD及CAT活力较照射对照组明显升高,其中照射对照组SOD、CAT活力分别为864±217、3.194±1.014;黄芪多糖灌胃组1821±181、7.334±1.022。三组LH及T水平未发现显著改变;
     结论
     APS可减轻大剂量60Coγ射线对睾丸间质细胞表面LHR的损伤,短期对睾丸间质细胞合成和分泌T的功能无明显影响。
Research background and objective:
     At present, people who are infertile in child-bearing age could be at a ratio of10%, male sterility accounts for about15%~50%in infertile couples. Study shows that there are a variety of factors can cause male spermatogenesis dysfunction (azoospermia, oligozoospermia and asthenozoospermia), such as no testosterone, radiation, bilateral undescended testicles, congenital seminiferous tubule dysplasia, viral orchitis caused by mumps, genital damage, local high temperature of testis. Along with the popularity of science and medical radiation therapy, the opportunity of contact rays is more and more, which leads to the number of people who have spermatogenesis dysfunction is increasing year by year.60Co-gamma rays radiation can cause testicular sperm cells, born support cells and stroma cells damage. Astragalus polysaccharides (APS) is dry powder extracted from the root of astragalus, which can be used as immune promoting agents or regulator, at the same time,it has the effects of anti-virus, anti-tumor, anti-aging, anti-radiation, anti-stress and antioxidant. Our study is designed to observe that after the great dose of60Co-gamma rays irradiating SD rats testicles locally, its pathological structure and the changes at molecular level. And by designing the control group, observing that whether common dose APS can reduce the damage caused by60Co-gamma rays.
     Methods
     Thirty male SD rats were equally randomized into three groups(n=10):the normal control group, the irradiate control group, the APS irrigation group. The structural changes of Leydig cells were observed by light microscope. The serum luteinizing hormone(LH)、testosterone(T) and catalase (CAT) levels were measured by ELISA; the Superoxide dismutase(SOD) measured by Hydroxylamine; and the mRNA expression levels of luteinizing hormone receptor(LHR) from Leydig cells surface were measured by RT-PCR.
     Results
     Compared with the normal control group,the Leydig cells exhibited size, cytoplasm, quantity decreases and the nuclear shrinkage after radiation. The mRNA expression levels of LHR were lower(p<0.01); in the APS irrigation group, the pathological changes of Leydig cells were lightly. The mRNA expression levels were close to the normal; the activity of the serum SOD and CAT were increased obviously;and the serum LH and T levels were no significant change.
     Conclusion
     APS can reduce the impaiment of Leydig cells caused by the higher doses60Coγ gammal ray irridate in rat. It was no significant change in testosterone synthesis in short-terms.
