槲寄生抗实验性心律失常作用的研究

摘要
槲寄生为桑寄生科槲寄生属,槲寄生中含有多种活性成分,现已从中分离出挥发油、黄酮、有机酸等小分子化合物及多肽、蛋白多糖等高分子化合物。槲寄生作为一种常用中药,具有抗肿瘤、抗氧化、抗衰老、抗病毒、抗心律失常等多种药理作用。近来这些中药制剂在临床上应用颇广,特别是在心脑血管病中更有独特作用。本课题主要通过建立心律失常动物模型来探讨槲寄生的抗心律失常作用,以及对心脏的保护作用机制。为临床工作提供理论依据和实验基础,为心律失常治疗开拓一条新的途径。
     本课题分为4部分:(1)将小鼠移入浸有3~4 ml氯仿棉球的倒置1000 ml烧杯中,小鼠呼吸停止,立即取出,迅速连接心电图机,观察心室颤动发生的百分率。结果表明:槲寄生能够明显降低心室颤动发生的百分率(2)舌静脉注射氯化钡诱发大鼠心律失常,建立心律失常动物模型。观察心律失常的发生时间、持续时间。结果表明:槲寄生可明显延长动物心律失常发生时间,缩短持续时间。(3)舌静脉注射乌头碱诱发大鼠心律失常,建立心律失常动物模型。观察室性早搏(VP)的发生时间、室性心动过速(VT)、室颤(VF)的发生时间以及动物死亡数。结果表明:槲寄生能够明显延长VP、VT、VF的发生时间,减少动物死亡数。(4)结扎大鼠左冠状动脉,建立心肌缺血再灌注心律失常模型。观察VP发生数及VP、VT、VF的发生时间,心律失常持续时间和心率变化。结果表明:槲寄生能够明显减少VP发生数延长VP、VT、VF的发生时间,缩短心律失常持续时间及降低心率变化幅度。
     通过本课题的实验研究,将对槲寄生抗心律失常作用机制有更深入的认识,从而为治疗心律失常提供新的思路和方法。
Objective: Arrhythmia is a serious symptom of heart performance and one of the major causes of death, Viscum Coloratum research Antiarrhythmic effect and its mechanism.
     Methods: ventricular fibrillation induced by chloroform, barium chloride-induced arrhythmia, myocardial ischemia and reperfusion in rats and arrhythmia aconitine induced arrhythmia, and other models, through experimental observation of Viscum Coloratum on experimental pacemaker disorders role.
     Results: Viscum Coloratum can significantly confrontation chloroform in mice induced ventricular fibrillation and ventricular fibrillation occurred was significantly decreased (P <0.05, P <0.01) significantly delayed barium chloride-induced arrhythmia in time and shorten the duration of (P <0.05, P <0.01) to reduce coronary artery ligation in rats induced arrhythmia VP arrhythmia frequency and shorten the time (P <0.05, P <0.01) to delay rats induced by coronary artery ligation pacemaker mad VP, VT, VF a time before and after the heart rate and reduce the rate of change (P <0.05, P <0.01) to delay Aconitine premature ventricular contractions induced arrhythmias (VP), a room of Echocardiography speed (VT) and ventricular fibrillation (VF) in time and reduce animal deaths (P <0.05, P <0.01).
     Conclusion: Viscum Coloratum can be effective against chloroform in mice induced ventricular fibrillation, and to reduce the incidence of ventricular fibrillation in mice. Viscum Coloratum on barium chloride-induced arrhythmias in rats resist, delay arrhythmias in time to shorten the duration of arrhythmia. Viscum Coloratum on Myocardial Ischemia-reperfusion arrhythmia caused a confrontation, VP can reduce the frequency and rate of change in heart rate, shorten the duration of arrhythmia, delayed VP, VT, VF in the time. Viscum Coloratum on aconitine induced arrhythmias in rats resist, delay VP, VT, VF the occurrence of time, reducing animal deaths. To sum up, Viscum Coloratum for a wide range of experimental arrhythmias significantly confrontation role.
