阿司匹林对大鼠骨质疏松性骨折愈合过程影响的实验研究

摘要
目的探讨阿司匹林对去势大鼠骨质疏松性骨折愈合过程的影响。
     方法3月龄雌性Wistar大鼠60只,随机分成去势组50只,假手术组5只和空白组5只。3个月后,从去势组中随机选取5只大鼠与假手术组及空白组的大鼠一起测骨密度,验证去势大鼠骨质疏松模型。确认模型成功后,将去势组大鼠全部造成左侧股骨中点骨折,用克氏针髓内固定。再将制成骨折模型的50只大鼠,随机分成A组和B组(每组25只),A组给予阿司匹林灌胃,33mg/次,1次/日,B组给予等量生理盐水。两组分别于骨折术后2、4、8、12周各取5只大鼠,测定骨密度值,然后处死取骨痂,行骨痂组织学检查,8、12周行放射学观察。两组剩余的5只大鼠于12周时行左股骨生物力学测定。
     结果①骨密度值:A组各时间点均高于B组,差异有统计学意义;②组织学观察:HE染色,A组软骨内成骨过程较B组加快;③放射学观察:术后8周,A组骨折线模糊,B组骨折线仍较清晰;术后12周,A组骨折线消失,B组骨折线模糊;④生物力学测定:术后12周,A组左侧股骨的三点弯曲载荷高于B组,差异有统计学意义。
     结论阿司匹林能够促进去势大鼠骨质疏松性骨折愈合,增加骨质的骨密度和生物力学强度。
Objective To research the effects of aspirin on healing process of ovariectomized rats with osteoporotic fractures.
     Methods60female Wistar rats which were3months old were dividied randomly into3groups:ovariectomized group50, pseudooperation group5, blank group5. The bilateral ovaries of ovariectomized group were resected; we do operation on peudooperation group without resecting bilateral ovaries, while adipose tissue with the same weight of two ovries were removed; do nothing with blank groupt.3months later, in order to verify whether osteoporosis models were successed bone mineral density of5rats randomly selected from ovariectomized group were detected, so are the pseudooperation group and blank group. After the osteoporosis models were built, we broke the left femur of ovariectomized rats at the midpoint, then used Kirschner wire to fix them intramedullary. Then the fracture model of50rats were divided into group A and group B (each group of25only), group A were lavaged with aspirin,33mg/time,1time/day, group B were lavaged with corresponding saline. Bone mineral density were detected at2th,4th,8th,12th week (each time we take five rats from each group) then we took callus and do histological examination. Besides we do radiology observation at8th,12th week. The biomechanics determination were done on the rest five rats of each group at12th week.
     Results①Bone mineral density values:The value of group A are higher than that of the group B at each time point;②Histologic observation:HE dyed, cartilage formation of group A is accelerated compared with group B;③Radiology observation:fracture line of group A were fuzzy while fracture line of group B is still clear after8weeks; fracture line of group A were disappear while fracture lines of group B were blurred After12weeks;④The biomechanics determination:, the three bending load of left femur of group A was higher than group B, after12weeks, the difference was statistically significant.
     Conclusions Aspirin can promote the healing process of ovariectomized rats with osteoporotic fractures, increase the bone mineral density and biomechanical strength.
