灯笼草对小鼠消化道过敏反应的预防作用研究

摘要
目的:观察灯笼草对卵清蛋白致敏小鼠消化道过敏反应有无预防作用,探讨能够有效预防儿童食物过敏的药物。
     方法:选择昆明种雌性小鼠64只,随机分为预防(Y组)、治疗(T组)、激发(CH组)和对照(NS组)四个大组;应用卵清蛋白建立致敏小鼠肠道模型;为小鼠灌服不同浓度的灯笼草果实水提物;采取血液和小肠标本,检测各组小鼠血清ALT和IgE水平;对小肠标本进行HE和甲苯胺兰染色及免疫组化染色,观察小肠粘膜变化情况,并计算肥大细胞完整率,检测IL-5、IFN-y的阳性率;最后将预防组和其他各组的结果进行比较研究。
     结果:1.激发组小鼠IgE、ALT、IL-5和IFN-y含量均明显升高,肥大细胞完整率明显降低。2.高、中浓度灯笼草预防组的IgE、ALT、IL-5和IFN-y等观察指标,均明显低于激发组,差异具有显著性(P<0.01),低浓度组不明显;肥大细胞完整率明显高于激发组(P<0.01)。3.预防组的IgE、IL-5和IFN-γ水平在数值上优于同浓度治疗组,但差异无统计学意义(p>0.05)。4.各个观察指标在预防组与治疗组之间未见显著性差异(p>0.05)。
     结论1.高、中浓度灯笼草可明显缓解小鼠过敏引起的消化道炎症。2.预防组缓解小鼠消化道过敏性炎变的表现均较激发组显著,但与治疗组作用相似。3.灯笼草对食物过敏具有明显地调节和缓解作用,但不能完全预防小鼠的肠道过敏反应。4.灯笼草可能是用于预防和治疗儿童食物过敏的有效药物,具有较高的临床研究价值。
[Objective] Whether there is the observation Physalis peruviana preventing the function to ovalbumen sensitization mouse digestive tract allergic reaction, the discussion can prevent the child food allergy effectively the medicine.
     [Methods] Choosed the Kunming female to plant mouse 64, divides into the prevention randomly (Y group), the treatment (T group), the stimulation (CH group) and the normal contral (NS group); Using ovalbumen establishment sensitization mouse digestive tract model; Aqueous extract of Physalis peruviana which filled the clothing different density for the mouse; Adopted the blood and the small intestine specimen, examined each group of mouse blood serum ALT and the IgE level; Carried on HE and the toluidine blue dyeing and the immunohistochemistry to the small intestine specimen, observed the small intestine mucous membrane changed situation, and calculates the mast cell percentage, examined IL-5, IFN-γmasculine gender rate; Finally we would compared with prevention group and other each group's result.
     [Results] 1.The stimulation group mouse IgE, ALT, IL-5 and IFN-γthe content obviously elevated, the mast cell percentage reduced obviously. 2. The high, middle Physalis peruviana prevention group's IgE, ALT, IL-5 and IFN-γobservation targets and so on, obviously is lower than the stimulate groups, there was the significance in the difference (P<0.05).3. The prevention group's IgE, ALT, IL-5 and IFN-γlevel and same density treation groups was different in value, but on signifincantly diversity (P>0.05).4.Each observation target could not watch the significance in the difference between prevention group and treation group(P>0.05).
     [Conclusion]1.High,middle density Physalis peruviana could alleviale the digestive tract inflammation which obviously the mouse allergy caused.
     2.In prevention groups,mouse digestive tract allergic inflammation changed the more remarkable alleviation than stimulates groups,but the treatment group function is simlla(?)
     3.Physalis peruviana had obviously the adjustment and the alleviation function to the food allergy, but could not prevent mouse's intestinal tract allergic reaction completely.
     4.Physalis peruviana possibly was used in preventing and controlling the effective medicine of the child food allergy,and had the high clinical research value.
