灯笼草对致敏小鼠肝损伤的保护作用

摘要
目的观察致敏小鼠是否确实存在肝损伤,探讨灯笼草果实及宿萼对卵清蛋白致敏小鼠肝损伤的影响,研究灯笼草防治小儿食物过敏更有效的药用部分。
     方法制备小鼠过敏模型;灌服不同浓度的灯笼草果实和宿萼的水提物;采取血液和肝脏标本,间接测定血清中IgE及ALT水平;并对肝脏样本进行HE染色及免疫组织化学染色,检测TNF-α、IFN-γ的阳性率;并对果实和宿萼组的药效作用进行比较研究。
     结果1.激发组小鼠IgE、ALT、TNF-α和IFN-γ含量均明显升高;2.高、中浓度灯笼草果实和宿萼,均可明显抑制卵清蛋白介导的小鼠被动过敏反应(P<0.01>,缓解小鼠过敏引起的肝脏炎症;实验组小鼠的IgE、ALT、TNF-α和IFN-γ含量明显低于OVA激发组,差异有显著性(P<0.01),低浓度组不明显。3.宿萼组降低IgE、ALT、TNF-α和IFN-γ的作用明显优于果实组(P<0.05)。
     结论1.致敏小鼠除肠道、皮肤、呼吸道炎变外,确实存在明显的肝损伤,这种损伤可能是由IgE介导的速发型免疫反应,并且与细胞因子IFN-γ和TNF-α有密切关系。2.高、中浓度灯笼草果实及宿萼对致敏小鼠的免疫性肝损伤均有明显的保护作用。3.灯笼草宿萼较果实对致敏小鼠抗炎抗过敏及保护肝损伤的作用更加明显。4.灯笼草对治疗小儿食物过敏所致肝损伤可能具有一定的疗效。
Objective: To observation sends the sensitive ratwhether truly has the liver damage, to investigate theinfluence of physalis Peruviana L on OVA induced senstitiveliver injury. To approach the medicinal purposes part ofchildren food allergy.
     Methods: Preparation small rat allergy model. Fill thedifferent density the physalis Peruviana L's fruit and calyxshuitiwu. Adopting blood and liver and indirect determinationblood serum IgE and ALT level; liver pathological changesunder microscope were observed. Histochemistry dyeingexamines the TNF-α、IFN-γmasculine gender rate. And Calyxgroup's fuction conducts the comparison rearch to the fruit.
     Results: 1.The stimulations groups rat IgE、ALT、TNF-α、IFN-γObviously elevates content; 2.The high and middledoseage of might inhibit the dermato-allergic reaction of ratand mitigating the inflammation of intestine. Blood serumIgE and ALT in the model group were higher than in normalcontrol group (p<0.01) the quantity of TNF-α、IFN-γwere higher than in normal group(p<0.01). Which proved that OVAintravenously had already led to rat liver damage. Calyxgroup'change is more oboiously.
     Conclusions: 1) Outside sends the sensitive rat exceptthe intestinal tract, the skin, the respiratory tract inflammationchanges, truly has the obvious liver damage, this kind ofdamage is the fast hairstyle immune response which maybelies between by IgE leads, and the cell factor TNF-α、IFN-γhas the close relation. 2) The high and middledoseagesphysalis Peruviana L's fruit and Sue sends the sensitive rat'simmunity liver damage to the OVA to the obviously protectivefunction. 3) To compare with fruit and Calyx to the OVA withthe result that the anti-inflammation. Anti-allergy fuction ismore obviously. 4) The physalis Peruviana L has somepossibly curative effect of children food allergy.
