交泰丸方药的半仿生提取法实验研究

摘要
目的:为交泰丸剂改研究优选提取工艺;进一步验证半仿生提取法(简称SBE法)的科学性和实用性。方法:先以小檗碱、肉桂酸、总生物碱、挥发油、干浸膏含量为指标,用均匀设计优选出交泰丸方药的SBE法工艺条件;再将方药组合成2组,用半仿生提取法提取,提取液作指标成分的含量测定及甲醇精制液的薄层色谱、挥发油的气相色谱比较,优选出交泰丸方药SBE药材的最佳组合方式;然后对该方药最佳组合的SBE液作不同醇沉浓度的成分比较,优选出半仿生提取醇沉法(SBAE法)的最佳醇沉浓度;在此基础上作SBE法、水提取法(WE法)、SBAE法、水提取醇沉法(WAE法)4种方法提取液的成分比较及SBE液与WE液的镇静作用比较。结果:交泰丸方药SBE的工艺条件为:第1煎用水以HCL调至pH2.20,第2、3煎用水依次以NaOH调至pH6.50,8.70;煎煮时间依次为170min,80min,40min;方药提取时以黄连、肉桂分别提取为佳;4种方法提取液5个指标成分的综合评价值为:SBE液>WE液>SBAE液>WAE液;SBE液较WE液的镇静作用明显增强。结论:交泰丸方药成分提取时以SBE法为佳。本研究进一步验证了SBE法的科学性和实用性,完善了中药复方剂改用SBE法的基本研究模式,并为中药药对的研究提供了借鉴。
Objective: Optimizing the extract technology in the modification of the formula of Jiaotai Pill and further verifying the correctness and practicality of semi-bionic extraction(SBE). Methods: At first, optimizing the SBE technical conditions for Jiaotai Pill ingredients through even design while berberine, cinnamic acid, total alkaloids, volatile oil and dry extract were adopted as indexs; Then, dividing the ingredients into 2 combinations and extracting by the SBE method respectively, determining the contents of index components in the each extract, comparing the TLC of absolute methanol solution and the GC of volatile oil, and making out the SBE s best ingredients combination For the formula of Jiaotai Pill. The SBE extract of the optimized ingredients combination was put to further comparison of components in different conditions of alcohol precipitating concentrations, confirming the most ideal alcohol precipitating concentration of the semi-bionic extraction by precipitation with alcohol (SBAE) for the formula. On this base, comparison of components were conducted over the four extracts derived from four different kinds of extraction methods that are the SBE, the water extraction(WE), SBAE, and the water extraction by precipitation with alcohol (WAE), and the mitigtive functions of the extracts of SBE and WE were compared. Results: The technical condition of SBE for the formula of Jiaotai Pill was: The pH value in first extraction was adjusted to 2.20 by the use of HCL solution. And then the pH values in the second and third extraction should be adjusted to 6. 50 and 8.70 by the use of NaOH solution, the durations of
    
    
    
    each section were 170min, 80min and 40min, respectively. it is better to extract Rhizoma Coptidis and Cortex Cinnamomi seperately. The comprehensive values assessed by the contents of 5 index components in the four extracts were : SBE >WE>SBAE>WAE. The mitigtive function of SBE extract is obviously more effective than that of WE. Conclusion: SBE method is the best among the four methods in the extraction of Jiaotai Pill components. The study firmly verifies that the SBE method is correct and practical, consummates the basic study model of Traditional Medicine compound s modification by using the SBE method , provides some experiences for Traditional Medicine s matching study.
