麻杏芎葶合剂对支气管哮喘大鼠肺组织PKC、MMP-9影响的实验研究

摘要
第一部分:动物实验:实验目的:观察中药麻杏芎葶合剂对支气管哮喘大鼠肺组织蛋白激酶C(PKC)、基质金属蛋白酶-9(MMP-9)表达水平的影响,探讨麻杏芎葶合剂治疗哮喘大鼠气道炎症的作用机制。
     方法:取SD大鼠60只,随机分为空白对照组、哮喘模型组、麻杏芎葶合剂低、中、高剂量组、地塞米松组。除空白组外,其他5组均行哮喘造模,麻杏芎葶合剂不同剂量组分别给予相应剂量的药液灌胃,模型组给予等量生理盐水,地塞米松组给予相应剂量地塞米松腹腔注射。采用免疫组织化学法观察各组大鼠肺组织PKC及MMP-9表达的变化,并光镜观察肺组织病理变化。
     结果:麻杏芎葶合剂可能通过抑制哮喘大鼠PKC及MMP-9的表达抑制炎性细胞的分泌,从而达到调节Th1/Th2细胞因子平衡,减轻气道炎症,干预气道重塑,缓解哮喘病情。

Abstract
     PartⅠ:Animal experiments:
     Objective:To observe the expression of Ma Xing Xiong Ting mixture of PKC and MMP-9 in bronchial asthma lung tissue and explore the Ma Xing Xiong Ting Continuous Mixture on airway inflammation in asthmatic rats and its mechanism study.
     Methods:SD 60 rats were randomly divided into control group, asthma group, the Ma Xing Xiong Ting mixture of low, medium and high dose group, dexamethasone group. In addition to the control group, the other five groups underwent asthma models, Ma Xing Xiong Ting mixture three groups were given different doses of the appropriate dose of liquid medicine orally, the model group were given normal saline and dexamethasone groups were given the same dose of dexamethasone intraperitoneal injection.Observing by immunohisto chemistry the expression of the PKC and MMP-9 in lung tissue of rats and the changes of lung tissue pathology by light microscopy.
     Results:Ma Xing Xiong Ting mixture may inhibit secretion of inflammatory cells by inhibitting the expression of PKC and MMP-9 in asthmatic rats, So as to achieve regulating Th1/Th2 cytokine balance to reduce airway inflammation and airway remodeling interventions and alleviate the asthma situation.
