偶氮染料与环糊精、DNA和蛋白质相互作用的研究及其分析应用

摘要
第一章 分别概述了染料、超分子化学、环糊精、脱氧核糖核酸(DNA)和蛋白质的基本内容,综述了近年来染料与环糊精、小分子与DNA及其小分子与蛋白质相互作用的研究进展。第二章 研究了偶氮染料活性艳红K-2G的电化学行为,建立了它的电分析方法。在0.10mol/L的NaCl(pH 6.9)底液中活性艳红K-2G有一稳定、灵敏的还原峰,峰电位约为—0.78V(VS.SCE),其峰电流与浓度在7.0×10~(-8)~1.0×10~(-4)mol/L范围内呈现良好的线性关系,检出限为6.0×10~(-8)mol/L。精密度、标准回收率均满足定量分析的要求。此外讨论了该染料分子还原电流的性质,探讨了电极反应机理。第三章 在底液为0.10mol/L的NaCl溶液中研究了偶氮染料活性艳红K-2G的极谱伏安行为。峰电位约为—0.78V(VS.SCE)处该染料有一稳定的、灵敏的还原峰,用线性扫描二阶导数极谱法研究了该染料与不同环糊精的相互作用。采用“电流法”测定了包结常数,比较了不同类型环糊精的包结能力,初探了包结点的可能位置,并对其进行了理论分析。第四章 研究了偶氮染料活性艳红K-2BP在0.10mol/L的NaCl(pH 6.9)底液中的极谱伏安行为。活性艳红K-2BP有一稳定的、灵敏的还原峰,以线性扫描极谱法研究了该染料与环糊精的相互作用。采用“直线法”测定了该染料与各种环糊精的包结比和包结常数,比较了不同类型环糊精的包结能力,初探了影响包结能力的可能因素。第五章 研究了偶氮染料碱性棕G的极谱伏安行为并建立了其极谱分析方法。结果表明,在pH 9.1的NH_4Cl-NH_3底液中,该染料产生一峰形好、稳定、灵敏的还原峰,其峰电位约为—0.67V(vs.SCE),峰电流与染料浓度在3.0×10~(-8)mol/L~1.0×10~(-4)mol/L呈良好的线性关系,检出限为9.0×10~(-9)mol/L。考察了16种染料对其定量检测的干扰,结果表明该法具有灵敏度高、线性范围宽、操作简便等优点,适于碱性棕G的定量分
CHAPTER 1 The fundamental contents of dye, supramolecule chemistry, cyclodextrin, DNA and protein were introduced. The research advances of the interaction of dyes with cyclodextrin, DNA and protein in recent years were summarized in details.
    CHAPTER 2 Electrochemical behavior of Reactive Brilliant Red K-2G was studied and a polarographic method for the determination of its have been established. A stable and sensitive linear-sweep voltammetric peak is obtained at -0.78V(vs.SCE) in NaCl (pH6.9) solution. The peak current is proportional to concentration over the range of 7.0× 10~(-8)~1.0×10~(-4)mol/L and detection limit is 6.0×10~(-8)mol/L.This method is suitable to apply for Reactive Brilliant Red K-2G of the quantitative analysis. The characteristic of the reduction peak current has been studied. At last the mechanism of electrode reaction was preliminary discussion.
    CHAPTER 3 Polarographic and voltammetric behaviors of Reactive Brilliant Red K-2G were studied. The result shows that Reactive Brilliant Red K-2G has a stable and sensitive reduction peak at potential of about -0.78V (vs.SCE) in the NaCl solution. Interaction of Reactive Brilliant Red K-2G with cyclodextrins was also studied by linear sweep second order derivative polarography. The corresponding inclusion constants were determined by "current method". Inclusion ability between the dye and different kinds of cyclodextrins was compared. A possible inclusion position was preliminary discussed and its theoretical analysis was given.
