持续光照对雌性大鼠生殖内分泌节律的影响及滋阴补阳序贯中药干预作用的实验研究

摘要
目的:探索持续光照对雌性大鼠生殖内分泌节律的影响及滋阴补阳序贯中药的干预作用。方法:选用42只3周离乳SD雌性大鼠,采用持续光照大鼠模型,随机分为光照组3组并与空白组对照,观察大鼠阴门开启情况。确定为阴门开启的大鼠,于阴门开启日称量体重。对阴道口开放的大鼠,自开启日起,每天上年8:00进行阴道涂片,观察出现第一个动情间期的时间,记录出现第一个动情间期日的大鼠体重。每周测定大鼠的体重,直至处死周。持续光照4周后,于凌晨2:00、14:00进行open field test。将光照组随机分为三组,分为持续光照+中药组,正常光照+生理盐水组,及持续光照+生理盐水组。连续用药18日后于凌晨2:00、14:00进行open field test。筛选动情前期大鼠颈动脉取血检测褪黑素及性腺轴激素。断头处死后手术摘取大鼠的子宫、卵巢,计算卵巢指数与子宫指数。结果:研究表明滋阴补阳序贯中药能使持续光照大鼠体质量上升;水平运动及垂直运动减少。能使持续光照大鼠阴道脱落细胞发生改变,缩短动情期,恢复动情周期;卵巢系数增高。滋阴补阳序贯方能使下降的MT和升高的T、E2、LH、PRL趋于正常。结论:研究表明持续光照能使雌性大鼠性启动提前,体质量下降、活动增多;持续光照能改变动情周期,延长动情期,缩短动情周期,使卵巢系数下降,持续光照能使MT水平下降(P<0.05),T、E2、LH、PRL均上升;滋阴补阳序贯中药可使雌性大鼠体质量恢复、水平运动及垂直运动减少。滋阴补阳序贯中药可使动情周期恢复,并能提高卵巢系数滋阴补阳序贯中药可使MT水平上升,使升高的T、E2、LH、PRL趋于正常。松果腺是参与下丘脑-垂体-性腺轴生殖内分泌调控的重要器官;滋阴补阳序贯调整生殖内分泌节律可能是通过松果腺实现的。
Objective:To explore the effects of female rats continusely exposed to light on reproductive endocrine rhythms and the functions of Chinese medicine with sequential interference by nourishing yin and invigorating yang. Choose42only three weeks weaning female SD rats, using the model that rats continuously exposed to light.The rats are randomly divided into3light groups,contrasting with blank group. Observe the opening situation of rat's vaginal orifice.Identified as the vulva opening rats,are measured the weight in vulva opening day. The vulva opening rats, since the opening day, every morning at8:00by vaginal epithelium smearing, are observed to the time of appearing first diestrus.Record the rats' weight of that time.The rats are determined the weight of rats weekly, until the week the rats to be put to death.After constant light for4weeks,the rats are taken open field test at2:00and14:00.The light group is randomly divided into three groups:continuous light+Chinese medicine group, normal light+NS group, and constant light+NS group.After continuous medication for18days at2:00,14:00,rats are taken open field test.The rats in proestrogen are selected to measur serum levels of melatonin and sexual hormone from the blood taken by their carotid artery.The rats are put to death with decollation, and gathered their uterine and ovarian through surgery,calculated the ovarian index and the uterine index.
     Results:Research shows that functions of sequential Chinese medicine by nourishing yin and invigorating yang can make continuous light rats' body quality rise; and reduce the horizontal movement and vertical movement.It can change the continuous light rats' vaginal exfoliated cells, shorten the estrogen period, recovery estrogen cycle; increase ovarian index. Functions of Nourishing yin and invigorating yang sequential can decrease the level of MT and make these increased T, E2, LH, PRL tend to be normal.
     Conclusion:The study shows the continuous light can make the female rats's sexual maturity earlier, reduce their body weight, increase activities; continuous light also can change the rats'estrous cycle, prolong diestrous phase, shorten estrous cycle,and make ovarian index decreased. Constant light can decrease the level of MT(P<0.05), T, E2, LH, PRL are all increased; nourishing yin and invigorating yang sequential Chinese medicine can make the female rats' body weight recovery, reduce the number in horizontal and vertical motions.The sequential Chinese medicine of nourishing yin and invigorating yang can restore sequential estrous cycle, and improve the ovarian index. It can also rise the level of serum MT, make T, E2, LH, PRL tends to be normal.The pineal gland is a vital organ involved in the hypothalamus-pituitary-gonadal axis in reproductive endocrine regulation;functions of nourishing yin and invigorating yang sequential adjustment on reproductive endocrine rhythms may be realized through the pineal.
