口蹄疫正向和反向间接血凝诊断液的研究

摘要
本试验应用从内蒙古地区分离的牛O型口蹄疫病毒株,制成佐剂抗原用其免疫动物,获得高免血清,通过硫酸铵盐析、凝胶层析的方法,对高免血清进行提纯,致敏绵羊红细胞,制成反向间接血凝诊断液。将口蹄疫病毒通过聚乙二醇浓缩、蔗糖密度梯度离心提纯后致敏绵羊红细胞,制成正向间接血凝诊断液。
    本试验进行了诊断液活力和特异性的检查,结果表明:正向间接血凝诊断液特异性好,只与同型阳性血清发生凝集反应。活力也比较高。达到1:512以上可以用其进行口蹄疫免疫后抗体检测。采用间接血凝法将制成的正向诊断液与兰州兽医研究所生产的诊断液进行对比,经过数据分析结果表明,利用内蒙古地区分离的口蹄疫病毒制成的正向间接血凝诊断液比已有的诊断液灵敏度高,且比较稳定。可以用来进行口蹄疫免疫后抗体检测。制成的反向间接血凝诊断液特异性好,但活力较差,虽然达到要求的标准但在进行对比试验检测口蹄疫抗原时,血凝效价结果低于兰州兽医研究所制造的诊断液。
A strain of O type FMDV isolated from Inner Mongolia district was used in this study. After being consentrated and purified, the virus was used to immune animals and high-immunify-level serum was obtained ; Then reverse indirect hemagglutination diagnostic preparation was obtained after sheep red blood cell was sensitized with the purifying the high-immunify-level serum by salt-outing and chromatographying. The indirect hemagglutination diagnostic preparation was obtained by sensitization of sheep red blood cell with purified FMDV
    The activity and speciality of the diagnostic preparation were tested . The result showed that the activity and speciality of the reverse indirect hemagglutination diagnostic preparation is good. It could only agglutinate the same type positive antibody and could be used to detect FMD antibody . The result of comparision of sensitivity between our diagnostic preparation and the diagnostic preparation produce by Lan Zhou Vererinary Institute showed that our diagnostic preparation had a higher and more stable sensitivity than the reverse indirect agglutination diagnostic preparation of FMD produce by Lan Zhou Vererinary Institut. Therefore , it can be used to FMD antibody detection . The speciality of indirect hemagglutination diagnostic preparation is good, but the result of comparision of sensitivity of our diagnostic preparation is lower than the diagnostic preparation produces by Lan Zhou Vererinary Institute.
