树鼩(Tupaia belangeri)血液生化学指标测定分析及HCV感染树鼩初步研究

摘要
树鼩(Tupaia belangeri)在医学生物学领域被广泛应用,但目前仍未实现其大量繁殖,国内外实验用树鼩仍然主要取自野外,树鼩实验前的很多背景都不清楚(如年龄、遗传学、寄生虫学、及自身携带疾病情况等),这样就无法保证实验数据的可靠性,实现其实验动物标准化是攻克上述问题的关键,建立树鼩的血液生化指标的正常参考值是其标准化建设的重要内容之一,本文建立人工饲养树鼩的血液生化指标的正常值范围,用于筛选合格实验动物和为诊断树鼩疾病提供依据,树鼩是研究人类疾病良好的替代者,本文为建立一种更有效、更稳定的树鼩丙肝模型进行一些有意义也有挑战的尝试。
     第一部分目的建立树鼩血液学和血液生化指标的正常值参考范围。方法:应用全自动血细胞仪和生化分析仪测定140只健康成年树鼩的血液学及血液生化指标。结果:雌性与雄性比较,LYM%、PLT、RDW%、GLU、CREA的差异具有显著性(P<0.05),TP、CHOL、TG的差异具有极显著性(P<0.01),其他指标的差异不具显著性。结论:本文建立了健康树鼩的血液学及生化指标的正常值参考范围,可为应用该动物进行科学研究时提供参考。
     第二部分目的探讨树鼩感染重组丙型肝炎病毒的可能性。方法本实验设立重组HCV病毒组(A组)、人阳性血清组(B组)、空白对照组(C组),通过巢式PCR法检测感染树鼩血浆的HCVRNA,检测树鼩血浆ALT。结果A组树鼩中5只树鼩在接种病毒后,血浆(分别在接种后第8天和第15天)中检测出HCVRNA,A组所有树鼩在接种病毒后均表现出ALT异常地升高,多数在第29天时达到峰值,B组树鼩中多数树鼩ALT出现异常升高,但未检测出HCVRNA。结论树鼩可以感染重组HCV,但只出现过一过性病毒血症,可能原因是HCV毒株的感染率及标准化、树鼩的个体差异及标准化等问题所致,树鼩能否替代黑猩猩成为HCV动物模型有待进一步深入研究。
Tree shrew(Tupaia belangeri) is widely used in medical and biological sphere.Tree shrew's production is not achieved,so the tree shrews for research still derive from wild catching mainly these days,and their background are unclear before tree shrews are used in experiment including age,genetics,parasitology and autologous disease,ect.And the reliability of experimental datas is unguaranteed.Achieving the standardization of experimental tree shrew is key to solve problems above.Meanwhile, establishing the reference of tree shrew's hematological and biochemical values is one important content of standardization.First,the normal reference of tree shrew's hematological and biochemical values in the thesis may be used to filter regular experimental tree shrew and to offer foundation of diagnosing tree shrew's disease;Second,the research is to carry out meaningful attempt to study HCV by using tree shrew.
     PartⅠObjective To establish and analyze the normal range of hematological and serum biochemical values in healthy tree shrew.Methods Blood samples were measured by automatic blood cellular analyzer and automatic blood biochemical analyzer.Results The reference of LYM%、PLT、RDW%、GLU、CREA were significantly different(P<0.05),and TP、CHOL and TG were obviously different(P<0.01) between female and male tree shrew. Conclusion The normal range of hematological and serum biochemical values in healthy tree shrew were established finally in the thesis,and these reference values in tree shrew are necessary for the scientific research using the tree shrew.
     PartⅡObjective To explore the possibility of tree shrew communicating HCV.Method The experiment is including p HCV team、human blood serum team and blank control.The HCVRNA of the tree shrews is checked by nested-PCR and ALT of blood plasm is examined.Result Five in A team with p HCV is checked HCVRNA+(eighth and fifteenth day) and the levels of their ALT increase abnormally in different degrees after ten tree shrews are inoculated p HCV,and the peak value of ALT of majority of ten tree shrews occurs at the twenty-ninth day,at the same time,the ALT of majority of B team is also higher unconventionally,but no HCVRNA+.Conclusion Tree shrew may infect HCV,but only the transitional viremia virusemia occurs.The reason may be caused by the ratio of HCV-infecting and its standardization,or individual difference of tree shrews and their standardization and so on.There need to be more farther and deeper research whether tree shrew is animal model of HCV instead of chimpanzee or not.
