生脉膨化胶囊对心血管疾病及免疫调节影响的实验研究

摘要
目的:生脉膨化胶囊是红参经膨化提取后研制出的新型胶囊制剂。观察生脉膨化胶囊对免疫抑制、心肌缺氧、心气虚小鼠及慢性心力衰竭大鼠的保护作用,研究其对心血管疾病及免疫调节的影响。
     方法:建立小鼠心气虚动物模型,检测小鼠心肌匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)、乳酸脱氢酶(LDH)、肌酸激酶(CK)含量;建立小鼠心肌缺氧动物模型,观察小鼠气管夹闭心电消失时间,特异性密闭耐缺氧小鼠存活时间;建立大鼠慢性心力衰竭动物模型,检测心脏重量指数(HW/BW)、左心室重量指数(LVW/BW)及血流动力学参数;建立小鼠免疫力低下动物模型,检测脏器指数、ConA诱导的小鼠脾淋巴细胞增值能力及骨髓DNA含量。
     结果:(1)生脉膨化胶囊可明显提高心气虚小鼠心肌匀浆中SOD活性,尤以生脉膨化胶囊中剂量组最为显著(P<0.01);可显著降低MDA含量,且中剂量组效果明显优于传统生脉胶囊组(P<0.01);能够显著降低心气虚小鼠心肌匀浆中CK, LDH酶活性(P<0.01),降低LDH的效果显著优于传统生脉胶囊(P<0.05)。
     (2)生脉膨化胶囊各剂量组脏器指数均高于正常组,中剂量组胸腺、脾脏、肾脏指数明显高于正常组(P<0.01),肝脏指数高于正常组(P<0.05);高中剂量组能显著提升免疫抑制小鼠的淋巴细胞增值反应(P<0.01);各剂量组DNA含量明显高于模型组(P<0.01),高剂量组高于传统生脉胶囊组。
     (3)传统生脉胶囊组及膨化胶囊各剂量组均可延长小鼠气管夹闭心电消失时间(P<0.01),且膨化胶囊各剂量组效果优于传统生脉胶囊组(P<0.05),高剂量组优于中低剂量组。膨化胶囊高中剂量组均可明显延长密闭耐缺氧小鼠存活时间(P<0.05),且效果显著优于传统生脉胶囊(P<0.05)。
     (4)生脉膨化胶囊能明显改善模型大鼠心衰症状、降低左心室指数(LVMI),减轻其组织病理性损害。生脉膨化胶囊治疗后均能明显升高±dp/dtmax及LVSP,并能明显降低LVEDP。
     结论:生脉膨化胶囊对心气虚小鼠具有保护作用,对环磷酰胺引起的免疫器官及主要脏器的变化、脾脏功能下降及骨髓损伤具有一定的保护和恢复作用;可延长小鼠平均存活时间,降低心肌耗氧量,具有显著的抗心肌缺氧作用;能改善慢性心力衰竭左心收缩和舒张功能,具有抗慢性心力衰竭的疗效;生脉膨化胶囊药效优于传统生脉胶囊。
Objective:Expanded Sheng-mai Capsule is a new type of preparation with expanded red ginseng for processing. Observed the protective effect on immunosuppression,myocardial hypoxia,Heart-Qi Deficiency and chronic heart failure in rats. Studied the influence of cardioascular disease and immunoregulation on Expanded Sheng-mai Capsule.
     Method:Established the animal models of Heart-Qi Deficiency in mice,detected the content of SOD、MDA、LDH and CK in myocardial extracts.Established the animal models of myocardial hypoxia in mice,the clipping trachea time,disappearance time of ECG,and survival time in specific closed hypoxia tolerance of mice were observed.Established the animal models of chronic heart failure in rats,detected HW/BW、LVW/BW and hemodynamic parameters.Established the animal models of low immunity in mice,detected the viscera index、the spleen T lymphocyte reproductive activity induced by ConA and DNA content of marrow.
     Result:(1) Expanded Sheng-mai Capsule can obviously improve SOD activity in myocardial extracts of mice,especially the middle dose group (P<0.01);the efforts of MDA reduction in the middle dose group is superior to the traditional Sheng-mai Capsule(P<0.01);CK,LDH enzyme activity were significantly decreased in Heart-Qi Deficiency mice,the reduction of LDH is superior to the Traditional Sheng-mai Capsule (P<0.05).
     (2)the viscera index in different dose groups of Expanded Sheng-mai Capsule were all higher than control group,the thymus index、spleen index and renal index of middle dose group were obviously superior to that of control group (P<0.01),the liver index was higher than control group (P<0.05).The high dose group and middle dose group could significantly raise cell proliferative responses of immunosuppression in mice (P<0.01);DNA content in different dose groups were much higher than model group (P<0.01),the high dose group was higher than Traditional Sheng-mai Capsule.
     (3) Expanded Sheng-mai Capsule and Traditional Sheng-mai Capsule could both prolong the clipping trachea time and disappearance time of ECG (P<0.01),different dose groups of Expanded Sheng-mai Capsule were all better than Traditional Sheng-mai Capsule (P<0.05),the efforts of high dose group was much better than the middle dose group and low dose group.The high dose group and middle dose group of Expanded Sheng-mai Capsule could obviously prolong the survival time in closed hypoxia tolerance of mice (P<0.05),and were much better than that of Traditional Sheng-mai Capsule (P<0.05).
     (4) Expanded Shengmai capsules could obviously improve the symptoms of heart failure in rats,decrease LVMI,reduce its organization pathological injury.Expanded Sheng-mai Capsules treatment can obviously increase±dp/dtmax and LVSP,and decrease LVEDP obviously.
     Conelusion:Expanded Sheng-mai Capsules could protect Heart-Qi Deficiency in mice,it may play a role in protect and recover change in immune organs and main organs、the changes of spleen function and bone marrow injury caused by cyclophosphamide,the mean survival time of mice was prolonged,decrease myocardial oxygen consumption.It has remarkable effect to myocardium against hypoxia.Expanded Sheng-mai Capsules may improve CHF systolic and diastolic function of the left ventricular,and has the efficacy of anti-CHF.The efficacy of Expanded Sheng-mai Capsules is better than Traditional Sheng-mai Capsule.
