川牛膝醇提物降压作用机理的实验研究

摘要
原发性高血压病是一种严重影响人类健康和生活质量的疾病。本病的发病率今年来呈上升趋势,现代医学对本病的研究已较深入,但仍有很多存在争议和未研究的问题存在,大量学者正为之不懈努力。中医学对本病的深入认识和理解是伴随中西医结合而逐步实现的,但至今尚无公认的系统化理论。临床上存在很多见解,多根据辨证论治的原则来治疗此病,已积累了大量的经验并取得了可喜的成果。
     原发性高血压病机极为复杂,因此临床难于辨证或用药难于取效。导师盖国忠教授认为本病的病位在血脉,病机关键是气血失和,血随气逆。牛膝醇提物就是在此理论指导下完成的,已在临床应用多年,取得较好的临床效果。为了进一步阐明牛膝的降压效果及有关机制,予以客观的,科学的论证。本实验以原发性高血压大鼠(SHR)为研究对象,此模型为国际上公认的与人类原发性高血压病最为接近的动物模型。通过对SHR的血压、血清超氧化物歧化酶(SOD)、丙二醛(MDA)的含量的影响;肾组织转化生长因子(TGF-β1)蛋白表达的影响;对体外培养血管内皮细胞影响;对血浆前列环素(PGI2)浓度影响的实验研究。对大鼠胸主动脉内皮细胞相关mRNA的表达的影响的实验研究。
     实验结果显示:
     1、川牛膝醇提物能够降低自发性高血压大鼠(SHR)的血压;
     2、川牛膝醇提物能明显降低大鼠血清MDA含量同时显著升高SOD活性;
     3、川牛膝醇提物可明显降低肾组织细胞中TGF-β1蛋白的表达;
     4、能够调节SHR血管活性物质生成和释放,升高血浆PGI2含量;
     5、川牛膝药物血清有促进体外培养大鼠胸主动脉血管内皮细胞增值作用;
     6、能够有效预防和修复LPS对血管内皮细胞的损伤。
     7、牛膝血清高、中剂量组可明显影响大鼠胸主动脉内皮细胞相关mRNA的表达
     本研究探讨了川牛膝部分作用机理,为认识和研究原发性高血压病的理论与方药提供了一定的依据,我们期待着更深入的研究与探讨。
The primary hypertension is one kind of the disease affecting human being health and the quality of life gravely. The morbidity falling ill originally assumes an uptrend since the beginging of this year , the modern medicine research to the disease is already more thorough , still there are existence , large amount of scholar there existing dispute and the problem not having studied in many being that unremitting efforts but. Knowing and understanding traditional Chinese medical science to going deep into falling ill originally is to accompany integration of Chinese and western medicine but to come true step by step , there is no so far still but universally accepted systematization theory. Mount existence clinically many view, many disease coming to cure this the principle according to the diagnosis and treatment based on an overall analysis of the illness and the patient's condition, already have accumulated large amount of experience.and have got gratifying achievement.
     Primary hypertension machine is extremely complicated , therefore clinical medicine difficult to discriminate or use is difficult to take an effect. The faithful director Gai guo zhong professor thinks that the disease place falling ill originally is in a blood vessel in which the blood and qi circulate , pathogenesis key is that gas blood becomes estranged , blood goes against with the gas. The root of bidentate achyranthes formulating is completed under being that here theory guides, already in applying many years clinically, get fairly good clinical effect. Expound and prove for expounding the root of bidentate achyranthes decompression effect further and giving the objective's , science's in connection with mechanism. The experiment takes primary hypertension rat (SHR) as the animal model studying the marriage partner , this model are universally accepted go ahead between nations and the primary hypertension of human being approaches most. Pass the effect to SHR rat blood pressure , serum superoxide dismutase (SOD) , third dialdehyde (MDA) contents; Effect kidney organization conversion growth factor (TGF-β1) egg white is expressed; To extracorporeal culture blood vessel bast, the cell affects; The experiment that thickness affects to blood plasma PGI2 studies.
     The experiment bear fruit is demonstrated:
     1. the ethanol extracts from Cyathula officinalis Kuan is a thing being able to reduce spontaneity hypertensionrat (SHR) blood pressure;
     2.the ethanol extracts from Cyathula officinalis Kuan is a thing being able to reduce rat serum MDA contents at the same time notable power-ups SOD activity obviously;
     3. the ethanol extracts from Cyathula officinalis Kuan id a thing but reduces expressing of kidney organization cell middle TGF-β1 ;
     4. Being able to adjust SHR blood vessel active material coming into being and releasing , elevates blood plasma PGI2 contents;
     5.the ethanol extracts from Cyathula officinalis Kuan has promoting the extracorporeal culture rat chest aorta blood vessel bast cell added value effect;
     6.Being able to have an effect to take precautions against and restore the LPS loss to blood vessel
     7. In the root of bidentate achyranthes serum height, dosages may affect rat chest aorta bast cell relevance mRNA expressing obviously setthe ethanol extracts from Cyathula officinalis Kuan part effect mechanism having studied investigation and discussion originally, has provided the certain basis , our in expectation of more thorough research and investigation and discussion to the medicine knowing and studying primary hypertension theory and method.
