日粮粗蛋白质水平及添加尿素对肉鸡内源氨基酸损失量的影响

摘要
内源AA损失量(EAAL)是计算饲料AA真消化率的基础数据,其测值受多种因素影响。本试验应用简化一次注射法测定了强饲不同粗蛋白质(CP)水平日粮及添加0.1%尿素后肉鸡内源AA损失量。35只体重接近(1.5±0.08kg)的艾维茵肉仔鸡等分为七组,按每kg体重25g干物质(DM)强饲无氮日粮(NFD)、混合蛋白源的CP16%、CP18%、CP20%及CP16%+U(0.1%尿素)、CP18%+U、CP20%+U七种饲粮。强饲后立即皮下注射30uci/kg体重L-[4,5-~3H]Leu。于注射后第24hr采集每只鸡翅静脉血液2.0ml,用三氯乙酸沉淀蛋白,制成去蛋白血浆样。连续无损收集每只鸡48hr内排泄物。测定饲粮、排泄物风干样中AA、CP含量及去蛋白血浆样中游离AA含量。测定排泄物及血浆样中放射性强度。根据一次注射法原理计算各组日粮时内源Leu损失量,然后利用NFD模式推算其他AA内源损失量。计算各组日粮AA及CP表观消化率、真消化率。
     试验结果如下:
     1 日粮CP水平(16%、18%、20%)对肉鸡EAAL无显著影响(P>0.05)。
     NFD测得肉鸡内源∑AA为13666.40mg/kg DMI 48hr,实际含氮饲粮CP为16%、18%、20%时,简化一次注射法所测得内源∑AA分别为19503.21、16253.85、19529.10mg/kg DMI 48hr,较NFD时明显上升(P=0.057-0.075)。三个蛋白质水平之间EAAL无显著差异(P>0.05),但CP18%时EAAL较CP16%和CP20%时有下降趋势(下降16.7%)。
     2 不同CP水平时添加0.1%尿素对肉鸡EAAL无显著影响(P>0.05)。
     在日粮CP16%、18%、20%时添加0.1%尿素,肉鸡内源∑AA分别为18545.13、18147.08、18977.26mg/kg DMI 48hr,相对于未添加时19503.21、16253.85、19529.10mg/kg DMI 48hr无显著变化(P>0.05)。
     3 日粮CP水平为16%、18%及20%时,AA表观消化率呈二次变化曲线,以CP18%时最高(其中Ile、Lys、Pro、Ser、Colu和∑AA、∑NEAA差异显著,P<0.05);AA真消化率呈直线下降趋势(P>0.05)。添加0.1%尿素会降低三种CP水平时AA表观和真消化率(P=0.045~0.080)。
The endogenous amino acid losses(EAAL) of the broilers, which were force-fed various diets different in protein level and supplemented with 0.1% urea, were determined using the method of simplified Single Intravenous Injection of [3H] Leucine (SIIL).
    35 AV broilers (avg. 1.50kg body weight) were randomly divided into 7 groups, received seven kinds of diets respectively. The diets were nitrogen-free diet (NFD), diets with 16%, 18% ,20% crude protein (CP) and CP16+U (0.1% urea) , CP18+U, CP20+U. After force-fed in 25g DM per kg BW, 30u Ci[3H]Leu/kgBW was injected subcutaneously, the excreta in the following 48hr were totally collected, and venous blood samples were taken at 24 hr. AA and CP content in diets ,excreta and blood sample were detected , radioactivity of excreta and non-protein-plasma were as well as. After the endogenous losses of Leu were determined by SIIL method , other AAs' endogenous losses could be got with NFD model, then calculated the apparent and true digestibility of AA and CP in various diets.
    The main results were as follows:
    1 The EAAL does not differ significantly (P>0.05) when broilers received diets different in CP levels. The endogenous losses of ?AA was 13666.40 mg/kg DMI 48hr when broilers fed NFD, and it increased significantly (P=0.045~0.057) when broilers fed CP16, CP18 and CP20 diets respectively, expressed as 19503.21, 16253.85, 19529.10 mg/kg DMI 48hr. EAAL was not significant (P>0.05) among three CP levels, but it had a decreased tendency in CP 18% diet (down 16.7% in numeric).
    2 The EAAL does not differ significantly (P>0.05) when broilers fed supplementation of 0.1% urea in different CP levels. The endogenous losses of ?AA were 18545.13 18147.08 18977.26 mg/kg DMI 48hr when CP16, CP18, CP20 diets adding 0.1% urea, respectively.
    
    
    3 The ADAA seems to be a square curve when broilers fed CP16, CP18 and CP20 diets, and the data were highest in CP18% diet. At the same time, the TDAA were decreased straightly (P>0.05). Supplementation of 0.1% urea in diets reduced the ADAA and TDAA (P=0.045~0.080).
