摘要
中药复方是中华民族智慧的结晶,对治疗疾病有着极大的优势。复方的各种药理活性正日益为国际社会普遍接受。直接从天然产物,尤其是从复方提取物中筛选某一类活性成分(混合物)是新药研发以仿制为主向创新为主的战略性转变策略之一,也是发挥传统医药资源优势,创新民族药的一条重要而有效的途径。
     中药复方往往由多种活性成分组成。多种活性成分分别作用于机体的不同器官和不同靶点,相互协同,从生理、病理上对机体进行调理,经整体调节而发挥复方的综合疗效。利用现代科学方法和先进的技术手段阐明中药复方治疗作用的物质基础及其生物效用机制,对阐明中医药理论内涵具有特别重要的意义。复方研究的落脚点是追求其优良的整体功效,同时也不能忽视对其单一化学成分和有效部位的研究。
     本论文主要研究一种抗肿瘤复方中的鸡血藤的化学成分。鸡血藤是一味沿用千年的活血化淤中药,具有多种药理活性,已为现代临床证实。本研究采用色谱分离和重结晶的方法,对鸡血藤乙醇提取物化学成分进行了较深入的研究。共分离得到十个化合物,经鉴定分别为:二十八烷、β-谷甾醇、正十六酸、二十二烷酸、豆甾醇、苯并呋喃-6-醇-3-酮、芒柄花素、4′,7-二甲氧基异黄酮、琥珀酸和十八烷二酸。其中正十六酸、二十八烷、二十二烷酸、苯并呋喃-6-醇-3-酮、4′,7-二甲氧基异黄酮和十八烷二酸,均为首次在鸡血藤中发现。经GC-MS分析,在Ⅳ-2中,发现了豆甾-5,22-双烯-3β-醇乙酸酯和胆甾-3-酮,环1,2-乙缩二醛;在Ⅶ-3中,发现了豆甾-3,5-二烯-7-酮;Ⅷ-4中发现了豆甾-4-烯-3-酮。这四种甾醇类化合物均是首次从鸡血藤中发现。
     这些研究成果对进一步研究鸡血藤复方,具有积极作用。
Being composed of many activity components,curative effect of Traditional Chinese Medicine Compound is mutual effect of every components .Those components respectively acts on different apparatus and target. Modem technology used to research of Traditional Chinese Medicine Compound,can illuminate the core of herbalist doctor theory.The root of Traditional Chinese Medicine Compound research is to optimize whole effect. Chemical components of single compound cannot be ignored.
    The research survey on Suberect Spatholobus Stem was reviewed in this thesis, including plant source, chemical constituents, pharmacological activities and clinic applications.
    The enthanol extraction of Suberect Spatholobus Stem is separated by column seperation
    methods and recrystallization. Ten compounds were isolated and identified by MS, which are
    Octacosane, beta-Sitosterol, n-Hexadecanoic acid, Docosanoic acid, Stigmasterol, Coumaran-
    6- ol-3- one, Formononetin, 4',7-dimethoxyisoflavone, Succinic acid, Octadecanedioic acid,
    Octacosane, n-Hexadecanoic acid, Docosanoic acid, Coumaran-6-ol-3-one and Octadecanedioic
    acid were isolated from this plant first time.
    Some components were analyzed by GC-MS. Four kind of sterols were first found, too. They are Cholestan-3-one, Cyclic- 1,2-ethanediyl acetal; Stigamasta-5,22-dien-3-ol, Acetate; Stigamasta-3,5-dien-7-one; Stigamast-4-en-3-one.
    These achievements will be helpful for the application of Suberect Spatholobus Stem.
