摘要
目的:观察针刺预处理对心肌缺血再灌注损伤大鼠c-fos蛋白表达的影响,评价针刺预处理效应。
方法:30只健康Wistar雄性大鼠,随机分为5组:正常对照组、假手术组、缺血再灌注组、缺血预处理组、针刺预处理组。心肌缺血再灌注前给予针刺进行预处理,采用“推管法”结扎大鼠冠状动脉左前降支建立大鼠心肌缺血再灌注损伤模型。HE染色观察组织形态学变化,免疫组织化学法检测c-fos蛋白表达。
结果:针刺“内关”“膻中”穴可下调心肌缺血再灌注损伤模型大鼠c-fos蛋白的表达。针刺预处理组与缺血再灌注组(IR)比较差异显著(P<0.05),与缺血预处理组比较(IP)无显著差异(P>0.05)。
结论:针刺预处理可下调c-fos蛋白表达,并可诱导心肌缺血耐受,减轻心肌缺血再灌注损伤程度,从而起到保护心肌的作用。
Objective: To investigate effect of acupuncture preconditioning on c-fos protein in myocardium of rat with myocardial ischemia reperfusion injury, to evaluate the effect of acupuncture Preconditioning
Methods: 30rats were divided into five groups randomly in the experiment: normal group,sham-operation group,ischemic group,ischemic preconditioning group, acupuncture preconditioning group. This thesis adopts needle prick vessel model rats before myocardial ischemia. Through using "pushing pipe"-tie off the rat coronary artery left anterior descending branch and establish the model of rat myocardial ischemia reperfusion. HE stains to observe change of histomorphology. Immunohistochemistry test c-fos protein expression.
Results: Acupuncture at "Nei Guan","Tan zhong " can reduce expression of c-fos protein in myocardium of rats with myocardial ischemia reperfusion injury. Acupuncture preconditioning group and ischemic reperfusion model group (IR)was significantly differenct (P <0.05), acupuncture preconditioning group compared with ischemic preconditioning (IP) was not significantly different (P> 0.05).
Conclusion: Acupuncture preconditioning can lower c-fos protein expression,induce ischemic tolerance,reduce myocardial ischemia reperfusion injury,played the role of myocardial protection.
