三高茶对实验性胰岛素抵抗综合征大鼠影响的初步研究

摘要
胰岛素抵抗综合征包括一组致动脉粥样硬化的症侯群:中心性肥胖,糖耐量降低/2型糖尿病,高胰岛素血症/胰岛素抵抗,血脂紊乱,高血压,脂肪肝和过早动脉硬化/冠心病等。胰岛素抵抗综合征的每一项都会增加心血管病的危险性,同时合并多种异常时发生心血管疾病的危险就更大。现在认为胰岛素抵抗综合征的病变过程大致是:胰岛素抵抗→高胰岛素血症→多种代谢和生理紊乱→动脉粥样硬化→心血管疾病。本课题就是通过研究保健食品“三高茶”对于实验性胰岛素抵抗综合征大鼠的多个心血管危险因素的干预作用,来探讨中医药在预防胰岛素抵抗综合征方面的作用。
     本论文由文献综述、理论探讨和实验研究三部分内容组成。
     文献综述部分中包括三篇综述:“胰岛素抵抗综合征研究进展”概述了现代医学对于胰岛素抵抗综合征的发病机制的认识、当前的诊断标准及治疗原则。“中医药防治胰岛素抵抗的临床研究进展”和“中医药防治胰岛素抵抗的实验研究进展”分别概述了中医药学对于防治胰岛素抵抗这一胰岛素抵抗综合征的中心环节的作用的现代实验和临床研究。
     理论探讨部分运用中医理论研究了胰岛素抵抗综合征的中医病机及治法,提出热毒内生、肝阳上亢、痰浊阻滞三个病理因素导致了胰岛素抵抗综合征并贯穿病变过程的始终。并从本草学和现代药理研究两个方面论述了三高茶的配方依据。
     在实验研究部分中主要探讨了三高茶在预防大鼠的实验性胰岛素抵抗综合征中的作用。在本实验中通过给予大鼠高脂高糖高盐膳食8周以诱发胰岛素抵抗及胰岛素抵抗综合征,同时对于实验组灌胃给予三高茶。实验结果显示,三高茶有助于改善大鼠的胰岛素敏感性指数及葡萄糖耐量,并能有效预防体重、血压、空腹血浆胰岛素水平、游离脂肪酸水平的升高和心脏、肝脏重量的增加并能对该模型的脂肪肝起到一定预防作用。
     本课题的研究结果显示,三高茶可有效地预防由热毒内生、肝阳上亢、痰浊阻滞三个病理因素引发的胰岛素抵抗综合征。作为中药对胰岛素抵抗综合征干预作用的初步研究,本课题为中医药防治胰岛素抵抗和胰岛素抵抗综合征提供了理论和实验依据。
This dissertation is composed of three parts: comprehensive review, theoretical study, and the experimental study of SanGao Tea on the preventing of insulin resistance syndrome.
    The first part is the comprehensive review on insulin resistance syndrome and insulin resistance, there are three summaries: the studies of insulin resistance syndrome by modern medical, the clinical observation and experimental research of herbs in treating insulin resistance, which is known as the main pathogeny of insulin resistance syndrome.
    The second part is the theoretical study on the insulin resistance syndrome and SanGao Tea of TCM, includes two essays: the comprehension on the pathogeny and cure of insulin resistance syndrome of TCM, the formula of SanGao Tea, including historical herbal lore and modern pharmacodynamics research about the herbs.
    In the third part, the part of experimental research, we try to explore the effects of SanGao Tea on preventing insulin resistance syndrome. In the experiment, rats were being given the high fat-sugar-salt diet for 8 weeks to build the model of insulin resistance and insulin resistance syndrome, and the SanGao Tea were given by the way of intragastric infusion from the beginning of the diet. The results indicate that SanGao Tea can improve the insulin action index and impaired glucose tolerance, and the SanGao Tea can prevent the increasing of blood pressure, the level of Plasma free fatty acid, fasting Plasma insulin, body weight, the weight of heart and liver.
    In summary, the whole study shows that SanGao Tea is an effective formula on the preventing of insulin resistance syndrome.
