川木香的化学成分及质量标准的初步研究

摘要
川木香为菊科川木香属植物川木香Vladimiria souliei(Franch.)Ling.及灰毛川木香Vladimiria souliei(Franch.)Ling vat.cinerea Ling.的干燥根。曾一度被作为为中药木香的地方习用、代用品,1963年开始被《中国药典》单独收载,直到2005年。主产于四川地区,为中医常用行气止痛药,同时也是藏族、羌族、蒙古等族习用药,主要用于消化道疾病。历史上木香类药材品种众多,名称几经变化,导致现今临床运用混乱。本课题从川木香本草考证出发,研究了川木香药材的来源、功效、炮制及现代研究,对其进行了化学成分研究。继而在化学成分研究结果的基础上进行了其标志性成分木香烃内酯和去氢木香内酯,以及分离得到的三个化合物的含量测定;并通过色谱指纹图谱,初步建立了川木香药材的质量标准。
     1木香类药材的本草考证
     通过查阅数量众多的古代文献,对木香类药材的品种和名称的历史变迁进行了完善的考察和整理,理清各种代用品、混淆品之间的关系。
     2川木香的化学成分研究
     采用硅胶柱层析方法,对川木香醇水双提物的乙酸乙酯部位进行了化学成分研究,共分离得到11个化合物,分别为α-amyrin margarate(Ⅰ)、Isobauerenyl acetate(异降香萜烯酸乙酸酯)(Ⅱ)、Dehydrocostus lactone(去氢木香内酯)(Ⅲ)、Uvaol(熊果醇)(Ⅳ)、15-Acetoxycostunolide(15-乙酰氧基木香烯内酯)(Ⅴ)、Santamarine(珊塔玛内酯)(Ⅵ)、Prinsepiol(青刺尖木脂醇)(Ⅶ)、Syringaresinol(丁香脂素)(Ⅷ)、VladinolD(川木香醇D)(Ⅸ)、Costunolide(木香烃内酯)(Ⅹ)、Cynaropicrin(菜蓟苦素;洋蓟内酯)(Ⅺ)。除去木香烃内酯、去氢木香内酯及川木香醇D为已知化合物外,其余均为初次在川木香药材中发现。
     3川木香生品及煨品的质量标准研究
     通过高效液相色谱法,用木香烃内酯、去氢木香内酯以及化学成分研究分离得到的3种化合物(化合物Ⅱ、Ⅷ、Ⅺ)作为对照品,对川木香进行了含量测定。分析结果不同产地川木香在此5个化合物含量上有一定区别;采用高效液相色谱法,以木香烃内酯为指标,对川木香药材进行了指纹图谱研究。
Radix Dolomiaeae is the dried rhizome of Vladimiria souliei(Franch.)Ling.and Vladimiria souliei(Franch.)Ling vat.cinerea Ling.It was used as a succedaneum of Radix Aucklandiae for a long time.1963,began to be“chinese pharmacopoeia”singal collection until 2005. It is mainly produced in Sichuan.It is uses to relieving Qi and relieve pain as Chinese traditional medicine,but also Tibetan, Qiang, Mongolia, habitual medicine, mainly for digestive diseases.There were many varieties of plants which were used as Mu-xiang in history,and the name changed several times,leading to the Clinical application of chaos. From the textual research of materia medica,members of this project investigated the resources、effect、processing,carried out its chemical constituents.Then assayed Costunolide、Dehydrocostus lactone and other three compounds which obtained by ourself by HPLC. Initially established its qu-ality standards by chromatographic fingerprint.
     1 materia medica herbal textual research of Mu-xiang and
     Through access to a large number of ancient literature,we did complete inspection and sorting on its species and names of the historical changes. Clarified the relationships between the various alternatives and the confused.
     2 Chemical constituents of Radix Dolomiaeae
     Silica gel column chromatography was used to analysis Radix Dolomiaeae.We got 11 compounds from it ,α-amyrin margarate(Ⅰ)、Isobauerenyl acetate(Ⅱ)、Dehydrocostus lactone(Ⅲ)、Uvaol(Ⅳ)、15-Acetoxycostunolide(Ⅴ)、Santamarine(Ⅵ)、Prinsepiol(Ⅶ)、Syringaresinol(Ⅷ)、VladinolD(Ⅸ)、Costunolide(Ⅹ)and Cynaropicrin(Ⅺ)。except Costunolide、VladinolD and Dehydrocostus lactone are known active ingredients,others are found in Radix Dolomiaeae the first time.
     3 Quality Standards of Radix Dolomiaeae
     By using the method of HPLC to determine the content of Dehydrocostus lactone、Costunolide、bauerenyl acetate、Syringaresinol and Cynaropicrin.The results showed that there are certain distinctions between the content of these compounds. By using the method of HPLC, cost-unolide be the index , we conducted fingerprint on Radix Dolomiaeae, established its quality standards initially.
