消劳眼舒颗粒对实验性树鼩肝劳模型瞳孔括约肌的影响

摘要
目的:观察消劳眼舒颗粒对实验性树鼩肝劳模型瞳孔扩约肌的影响,并探讨其作用机理。
     方法:以树鼩作为实验动物,对其限定照明时间并予以形觉刺激诱视,造成树鼩持续视近状态以模拟肝劳的病理变化。实验树鼩随机被分为6组:正常对照组、模型组(阴性对照组)、低、中、高剂量消劳眼舒颗粒组和阳性对照组。以放射性配基结合分析法检测虹膜MAChR(muscarinic acetylcholine receptor毒蕈碱乙酰胆碱受体,简称M受体)的RT值(受体最大结合容量)和K_D值(平衡解离常数),并对瞳孔扩约肌行HE、Masson、a-SMA染色,采用Mias-2000图形分析系统对其进行分析。
     结果:(1)与正常对照组相比,模型组MAChR的K_D值明显降低,各治疗组MAChR的K_D值明显高于模型组(P<0.01,0.05);各组间MAChR的RT值无统计学意义(P>0.05)(2)与模型组相比,各治疗组瞳孔括约肌HE染色平均光密度明显降低(P<0.01,0.05),平均黑度无明显变化(P>0.05);Masson染色各组平均光密度及平均黑度均无明显变化(P>0.05);与模型组相比,各治疗组瞳孔括约肌α-SMA(α-smooth muscle action,α平滑肌肌动蛋白)染色平均光密度无明显变化(P>0.05),平均黑度明显降低(P<0.01,0.05)。
     结论:结合前期实验研究结果,近距离久视所致支配树鼩瞳孔扩约肌

    ·的副交感神经持续异常兴奋可能是肝劳发病的重要原因之一。而消劳眼舒颗
    粒能有效抑制其副交感神经的持续兴奋并能缓解瞳孔扩约肌的痉挛。
Object: To investigate the effect of xiaolaoyanshukeli(消劳眼舒颗粒) on the sphinctre muscle of Tree Shrew and discuss the mechanism.
    Methods:Tree Shrew was made staring continuously.They were divided into 6 groups randomly: nomal control group, model group (negative control group),positive control group , three xiaolaoyanshukeli groups , including high dose, middle dose and low dose. Iris of Tree Shrew was separated to quantitate the MAChR by radioimmunossay and the pathologic histology changes of sphinctre muscle were oberved by Mias-2000 image analysis system.
    Results: (1) Compared with the nomal control group, the Kd of MAChR in the sphinctre muscle of Tree Shrew in model group reduced (P <0.05).Compared with the model group, the Kd of MAChR in all therapy groups significantly improved (P<0.01,0.05). The RT of MAChR in all groups was not statistic value .(2) Compared with the model group ,the average optical density in HE staining of the iris sphinctre muscle in all traditional Chinese medicine therapy groups significantly decreased (P<0.01,0.05) .The average optical density and the average grey level in Masson staining of all groups were not statistic value; Compared with the model group , the average grey level in a-SMA staining of the iris sphinctre muscle in all therapy groups significantly decreased
    
    
    (P<0.01,0.05).
    Conclusion: The persistent erethism of parasympatheti controlling iris sphincter muscle may be one of important reasons of GanLao (肝劳) .Xiaolaoyanshukeli could adjust the excitability of parasympathetic nerve and relax contracting muscle.
