针刺对快速老化模型小白鼠SAMP6骨质疏松及微量元素含量影响的实验研究

摘要
本课题以自日本引进的自然快速老化模型小鼠SAMP6为实验对象,以正常老化的SAMR1小鼠为对照,观察其骨组织形态计量学变化,股骨及各脏器内微量元素含量情况,以及补肾法针刺“肾俞”对其的影响作用,探讨针刺对骨质疏松的影响作用。结果表明:
     1.通过骨形态计量学观察表明,与正常老化模型SAMR1相比,SAMP6小鼠自三月龄起就存在骨质疏松表现,与日本学者研究结果基本一致。证明SAMP6小鼠引进中国近10年来,在我院繁殖成功,其病理性状没有发生改变,是一种新的骨质疏松动物模型。
     2.用原子吸收法测得SAMP6小鼠股骨中Cu、Zn、Mg、Ca;脑内Cu、Hg;肝内Mg;皮肤Cu;血清Zn、Ca、Fe含量显著低于SAMR1;而心内Zn、Mg;肺内Cu、Fe;肝内Cu;肾内Cu、Zn;皮肤Zn、Mg;血清Cu、Mg显著高于SAMP1。快速老化小鼠SAMP6与正常老化小鼠体内微量元素水平及分布存在差异。造成这种差异的原因可能与其加速衰老和自发性骨质疏松有关。
     3.针刺“肾俞”后,SAMP6小鼠骨中Ca、Mg、Zn等的含量增加,血清Cu、肺中Fe含量显著降低,对于血清Ca和Fe以及其他器官微量元素含量有一定的影响作用。针刺对SAMP6小鼠体内微量元素水平有影响作用。
     4.针刺“肾俞”,SAMP6小鼠骨形态计量学参数有一定程度的改善。针刺对SAMP6骨质疏松可能有良性作用。
Object: To study the effects of acupuncture on bone histomophometry and trace elements concentrations of femur and viscera of SAMP6. Methods: The femur of 3-month-old, 6-month-oId SAMP6 and SAMR1; SAMP6 after acupuncture were processed undecalcified for bone histomophometry, compared with SAMP6 without acupuncture and SAMR1. Trace elements concentrations of femur and viscera were assessed by means of atomic absorption spectroscopy in SAMP6. SAMR1 and SAMP6 after acupuncture. Results: 1. SAMP6 mice show early-onset and progressive osteoporosis, compared with SAMR1 in bone histomophometry. 2. There is a difference in trace element level and distribution between SAMP6 and SAMR1 mice. There are significantly decreased in Cu, Zn. Mg and Ca concentrations of femur; Cu and Mg concentrations of brain: Mg concentration of liver; Cu concentration of skin; Zn, Ca and Fe concentrations of blood serum in SAMP6 mice compared with SAMR1, while significantly increased in Zn and Mg concentrations of heart; Cu and Fe concentr
    ations of lung; Cu concentrations of liver; Cu and Zn concentrations of kidney; Zn and Mg concentrations of skin; Cu and Mg concentrations of blood serum in SAMP6 mice compared with SAMR1. 3. After acupuncture "Shenshu" point, there are significantly increased in Ca, Mg and Zn concentrations of the femur, and significantly decreased in Cu concentration of serum; Fe concentration of lung in SAMP6 mice. 4. Acupuncture on "Shenshu" point can improve some bone histomophometry indexs to some degree. Conclusion: The results proved that SAMP6 mice were a novel osteoporosis animal model, which characteristic has no change since SAMP6 were introduced to China. It is successful breeding of SAMP6 in our hospital. It is possible that acupuncture has the function of regulating the level of trace element in S AMP6 mice and acupuncture maybe have a good effect on senile osteoporosis in SAMP6 mice.
