HMC毒素对玉米CMS与N细胞质线粒体结构和功能的影响

摘要
本研究用电镜技术观察C毒素处理同核的正常细胞质(N)和C型不育细胞质(C)玉米叶片的线粒体,结果表明,C毒素处理后的C细胞质玉米叶片的线粒体外膜完整,内膜受到破坏,嵴破坏严重,有的甚至看不到嵴。而同核的N细胞质玉米叶片的线粒体外膜完整,内膜只受轻微的破坏。未经C毒素处理的同核的N、C细胞质玉米叶片的线粒体内外膜均完整,嵴也很明显。
     用氧电极法测定C毒素对同核的N、C细胞质玉米叶片线粒体呼吸速率的影响,结果表明,加C毒素后,C细胞质玉米叶片的线粒体的呼吸速率下降等于或大于20%,呼吸抑制值的差等于或大于0.22;而N细胞质玉米叶片的线粒体的呼吸速率下降等于或小于5%,呼吸抑制值的差等于或小于0.06。
     从上述结果可以看出,C毒素对C细胞质玉米线粒体结构和功能的影响具有专化作用,且C毒素的作用位点可能在线粒体内膜上,但是这一推测还有待于在分子水平上的进一步研究。
The mitochondria from the normal cytoplasm (N) and CMS-C of Maize treated with C-toxin were observed by a transmission electron microscope (TEM) . The result showed that the outer membrane of C cytoplasmic mitochondria, after treated with C-toxin, appeared intact, the inner mitochondria! membranes were destroyed, and the cristae showed fragmentation to form small vesicles. But the outer membranes of N cytoplasmic mitochondria also appeared intact, and inner membranes were destroyed slightly. The outer and inner membranes of mitochondria not treated with C-toxin all appeared intact, and the cristae showed complete.
    The respiratory control ratios (RCR) were determined in the presence and absence of C-toxin with a Clark Oxygen electrode. The result showed that RCR declined equal to or more than 0.22 in the presence of C-toxin in C cytoplasm and respiratory ratio declined equal to or more than 20%; but in N cytoplasm RCR declined equal to or less than 0.06 in the presence of C-toxin and respiratory ratio declined equal to or less than 5%.
    From the above results, it is very clear that C-toxin is specificity to the structure and function of N and C cytoplasmic mitochondria. The site with C cytoplasm exists possible in the membrane of C cytoplasmic mitochondria. But the inference needs to study further in molecular level.
