针刺预适应对全脑缺血大鼠脑组织超微结构及细胞凋亡影响的实验研究

摘要
为了探索防治缺血性脑血管病的新途径,揭示针刺预防缺血性脑卒中的机理,本文以Wistar大鼠为受试对象,脑缺血前给予针刺,采用“4-动脉阻断”方法建立大鼠全脑缺血模型,脑缺血后大脑皮层、海马CA_1区HE染色观察组织形态学变化、神经细胞尼氏体亚甲蓝特殊染色法观察神经细胞损害,电镜观察组织超微结构,细胞凋亡原位末端标记(TUNEL染色)法检测神经细胞凋亡。实验结果显示:与正常组、假手术组相比,脑缺血组大脑皮层、海马CA_1区神经细胞缺失明显。与脑缺血组相比,脑缺血预适应组、手针预适应组、电针预适应组大脑皮层、海马CA_1区神经细胞损伤小,水肿程度轻,神经细胞形态恢复快,凋亡细胞数目少,电针预适应组与脑缺血预适应组相似,但优于手针预适应组。
     结论:针刺预适应对全脑缺血大鼠大脑皮层、海马CA_1区神经细胞具有保护作用;针刺预适应可以抑制全脑缺血大鼠大脑皮层、海马CA_1区神经细胞凋亡;针刺可以作为一种脑缺血的预适应手段,可以诱导脑缺血耐受,减轻缺血后神经细胞损伤。
To explore new preventing and cure method of cerebrovascular disease and preventive mechanism on ischemic cerebral apoplexy of acupuncture, this thesis acupuncture before brain ischemia in four vessels occlusion(4-vo) model rats. After ischemia, in cerebral cortex and hippocampal CA1 section use HE staining to observe change of histomorphology, neuronal Nissl's staining to observe neuronal damage, TUNEL method to display apoptosis state, electron microscope to observe ultrastructure. Result :compared with normal group , sham-opration group, the lose neuron in cerebral cortex and hippocampal CA1 section of ischemic group is evident , apoptosis is more evident after 24h, 48h, 72h, compared with ischemic group ,neural damage , dropsy and apoptosis in BIP group , acupuncture preconditioning group and electro-acupuncture preconditioning group is little, recovery of neuron is rapid, electro-acupuncture preconditioning group is perior to acupuncture preconditioning group. Conclusion: pre-acupuncture have preventive effect on neuron of ischemic cerebral cortex and hippocampal CA1 section, pre-acupuncture can prevent apoptosis, pre-acupuncture can act as a method of BIP, can induce BIT and lighten neural damage of ischemic cerebrovascular disease.
