芎麻汤提取物药效活性部位筛选的实验研究

摘要
目的:采用目前较先进的闪式提取法,分别得芎麻汤(XMT)石油醚、二氯甲烷、乙酸乙酯、正丁醇提取物,研究XMT及其各提取物对偏头痛模型(硝酸甘油(NTG)型、利血平低五羟色胺(5-HT)型)行为学症状、体重、凝血时间、血液指标(5-HT、NO)等影响,初步筛选出XMT治疗偏头痛的药效活性部位,同时对此有效部位进行抗炎、镇痛实验研究。
     方法:利用不同极性有机溶剂对XMT进行梯度闪式提取,得XMT四种提取物;以NTG型大鼠为研究对象,大鼠挠头及爬笼次数、血浆中5-HT、NO为观测指标探讨XMT及其各提取物对模型大鼠的影响以利血平低5-HT型小鼠为研究对象,小鼠体重、凝血时间及血浆中5-HT、NO为观测指标,探讨XMT及其各提取物对模型小鼠的影响;以热板法实验观察XMT及有效部位的镇痛作用;以醋酸致小鼠腹腔毛细血管通透性实验观察XMT及有效部位的抗炎作用。
     结论:①XMT原药材饮片混合粉末372g,闪提得本研究试药石油醚提取物10.8g、二氯甲烷提取物8.1g、乙酸乙酯提取物3.4g、正丁醇提取物1.7g。②XMT及其正丁醇提取物对NTG型偏头痛大鼠的挠头次数有抑制作用;XMT对NTG型偏头痛大鼠的爬笼次数具有抑制作用(P<0.05)。③XMT及其正丁醇提取物能显著降低NTG型偏头痛大鼠血浆中5一HT、NO的含量(P<0.01)。④XMT及其正丁醇提取物能显著增加利血平低5-HT型偏头痛所致的小鼠体重,调节血浆中5-HT的过度降低(P<0.01);XMT及其乙酸乙酯、正丁醇提取物能延长偏头痛小鼠凝血时间,显著提高血浆中NO的含量(P<0.01)。⑤XMT及其乙酸乙酯、正丁醇提取物具有镇痛作用;⑥XMT及其正丁醇提取物具有抗炎作用。
Objective:Using the flash type extraction method, get the extract of Petroleum ether, Dichloromethane, Ethyl acetate and Butanol from Xiongma decoction respectively. And then study the Xiongma decoction and its extracts on migraine model (nitroglycerin type, reserpine low5-HT type) in behavioral symptoms, weight, and coagulation time, blood index, etc. preliminarily screen the Xiongma decoction's effective parts for migraines, and research its effective parts on the anti-inflammatory and analgesic experiments.
     Methods:Using different polarity organic solvent to extract Xiongma decoction, with time, number, solvent quantity for index, to reduce pressure concentration, and flick dry solvent, get the extract of Petroleum ether, Dichloromethane, Ethyl acetate and Butanol of Xiongma decoction; With NGT model rats for research object, and with the number of scratching and climbing, the plasma of5-HT and NO for observation index, To study the influence of Xiongma decoction and its extractson in the migrane rats model by nitroglycerin; With reserpine low5-HT model mouth for research object, and with weight, coagulation time and the plasma of5-HT and NO for observation index, To study the influence of Xiongma decoction and its extractson in the mouth model by reserpine; With hot plate experiment to study the Analgesic action of Xiongma decoction and its effective parts; With the experiment of inflammation model by acetic acid to study the anti-inflammatory of Xiongma decoction and its effective parts.
     Results:①To get the extract of Petroleum ether(10.8), Dichloromethane(8.1), Ethyl acetate(3.4) and Butanol(1.7) of Xiongma decoction from original medicine yinpian(372g).iongma decoction and tis Butanol extraction might have inhibition of the number of scratching in the migrane rats model by nitroglycerin; Xiongma decoction might have inhibition of the number of model rats'climbing(P <0.05).iongma decoction and its Butanol extraction could reduce the model rats'plasma of5-HT and NO significantly(P<0.01).iongma decoction and its its Butanol extraction could increase weight and prevent excessive decrease in5-HT(P<0.01); Xiongma decoction and its extracts of Ethyl acetate and Butanol could prolong coagulation time and improve the content of NO(P<0.01).iongma decoction and its extracts of Ethyl acetate and Butanol could have analgesic action.iongma decoction and its Butanol extraction could have anti-inflammatory action.
