电针干预骨癌痛大鼠的实验研究

摘要
目的:通过电针刺激乳腺癌骨转移大鼠,观察电针对骨癌痛的影响,讨论其可行性,并探讨电针治疗骨癌痛的作用机理,以指导临床运用
     方法:1、将24只体重180-220g的SD雌性大鼠,随机分为三组:空白组、假手术组、模型组。将Walker256大鼠乳腺癌细胞经腹水传代后(1×105cells)接种至SD大鼠左侧后肢胫骨骨髓腔内制备模型。与空白组、假手术组比较判断造模成败。2、将40只体重180-220g的SD雌性大鼠接种Walker256大鼠乳腺癌细胞造模后随机分为5组:空白组、模型组、电针组、药物组、针药组,每组8只,分别干预后,观察各组大鼠行为学、形态学及利用免疫组化法和Western-blot法观察抗酒石酸酸性磷酸酶(Tartrate resistant acid phosphatase,TRAP)、胶质纤维酸性蛋白(Glial Fibrillary Acidic Protein,GFAP)的表达。
     结果:
     ].大鼠接种Walker256乳腺癌细胞后,X线摄片可见术侧胫骨在接种后8天、14天和21天分别出现不同程度的骨质损伤和破坏双侧后肢热痛过敏,鼠尾出现机械性触痛过敏;术侧胫骨HE染色在接种后21天可见大量的肿瘤细胞;
     2.大鼠接种后21天胫骨重酒石酸酸性磷酸酶(TRAP)染色表达上调,同时激活脊髓胶质细胞的标志物GFAP;
     3.应用电针、唑来膦酸及电针+唑来膦酸治疗均能够使上调的TRAP表达下降,同时抑制被激活的GFAP;
     4.电针+唑来膦酸对骨癌痛的治疗作用明显优于单纯电针或唑来膦酸的作用。
     结论:
     1.利用Walker256乳腺癌细胞能够成功制出大鼠乳腺癌骨转移模型
     2.应用电针、药物及针药结合均可缓解大鼠骨癌痛,且针药结合疗效明显优于单纯电针或药物治疗;
     3.电针对癌症骨转移疼痛具有治疗作用,而且电针结合药物能够提高镇痛效果。
Objective:By means of electroacupuncture to the bone metastasis in breast cancer, observe the effects to the pain and compare the effects of electroacupuncture to the bone metastasis in breast cancer with of drugs.Discuss the feasibility of electroacupuncture to the bone metastasis in breast cancer and get the mechanism of electroacupuncture to the bone metastasis in breast cancer to direct clinical treatment.
     Methods:Twenty-four female SD rats weighed180to220gram were randomly divided into three groups.They are blank group, sham-operated group and model group.Cells of Walker256were injected into the hollow pith of left hind shinbone to establish the model. The cultured cells were transferred by ascites. The models were examined by comparing with blank group and sham-operated group.
     forty female SD rats weighted180to220gram are divided equally into fiver groups at random.They are blank group,model group, electroacupuncture group, drug group and electroacupuncture with drug group.Observe the rats'behavior and morphology. Observe the expression of TRAP and GFAP by the means of Western-blot and Immunohistochemistry.
     Results:Bone damage appear in the shinbone at8days,14days,21days after inoculation by X-ray. Both hind legs'heat pain and tails'mechanical tenderness were sensitive. A lot of tumour cells were found by hematoxylin-eosin staining at left hind shinbone, after inoculated21days.
     After inoculated21days, Rats TRAP was higher. GFAP was activating, which is the spinal glial cell marker.
     Electroacupuncture, Zoledronic Acid and electroacupuncture combined with Zoledronic Acid can make the high expression of TRAP low and restrain the activating GFAP.
     The effect of electroacupuncture combined with Zoledronic Acid is better than only with electroacupuncture or Zoledronic Acid.
     Conclusion:Animal models of human breast cancer bone metastasis can be established successfully by Walker256breast cancer cells.
     Electroacupuncture, Zoledronic Acid and electroacupuncture combined with Zoledronic Acid can help relieve the rats'pain of breast cancer bone metastasis. The effects of electroacupuncture combined with Zoledronic Acid were better than only with electroacupuncture or Zoledronic Acid.
     electroacupuncture can do more effective help to the pain of cancer bone metastasis and electroacupuncture combined with drugs can increase the effects.
