火麻种子水溶性多糖的提取、分离、纯化和化学结构的研究

摘要
火麻种子粉碎后经乙醇脱脂后用冷水抽提。经胃蛋白酶酶解后并联合Sevag法脱蛋白后,透析和冷冻干燥得粗多糖H。利用离子交换柱DEAE-sephadex A-50(Cl~-),琼脂糖凝胶Sepharose CL—2B和葡聚糖凝胶Sephadex G—100对粗多糖H进行分离纯化,得到两种精制多糖HS_1和HS_2,并对其进行了纯度鉴定,研究结果表明它们均为均一多糖组分。并用凝胶过滤法测定了两种多糖的分子量,分别为42795和15396。火麻多糖的热稳定性较差,高温或过酸和偏碱均会使其中的多糖部分将解,它在沸水中溶解性能较好,但不溶于高浓度的有机溶剂,多糖HS_1经过气相色谱分析,结果表明其单糖组成主要为D-阿拉伯糖,D-木糖,D-甘露糖,D-半乳糖和D-葡萄糖组成,其摩尔比为:1.5:1.2:1.6:2.0:3.0。用红外光谱初步分析了多糖HS~1和HS_2的结构,结果表明HS_1和HS_2都是β型糖苷键构型。
In this paper, Hemp seed was defat alcohol,water-extracting,pepsin-enzymolysis, dialysing,freezing and drying.At last,the water soluble polysaccharides H was obtained.Il was fractionated by gel filtration chrom atograph on DEAE-sephadex A-50(Cl-) and Sepharose CL-2B and Sephadex G-100,the results finded two kinds of polysaccharides HS1 and HS2. The results showed that they were all homogeneous porysaccharides.Furthmore,molecular weight was determined by gel filtration chrom atograph method,the result was 42795 and 15396. The polysaccharides of HS1 and HS2 would degrade under the condition of high temperature,acidity (ph<7=and alkalinity (ph>) .The HS1 and HS2 were soluble in hot water but insoluble in the high concentration solutions of such organic solvents as alcohol,ether,acetone,acetic ether etc. By the gas chromatography analysis of polysaccharides HS1 was testified that the main ingredients of monosaccharide compsing the HS1 are D-Arabinose, D-Xylose, D-Mannose, D-Galactose and D-Glucose and the mole rate of them are 1.5: 1.2: 1.6: 2.0: 3.0.I.R.SPECTRA suggested that both HS1 and HS2 have p-glycoside linkage.
