补阳还五汤抗脑缺血症神经细胞损伤的谱效学研究

摘要
补阳还五汤是治疗中医气虚血瘀证的经典良方,对治疗脑缺血、脑中风、血栓、血脂及改善免疫方面具有显著作用。该方成分众多,作用复杂,其有效性基础及作用机制一直未能阐明。若能建立适应中药复方特点的有效性研究方法,有望能为中药复方现代化研究提供新的思路与依据。
     目的:本文以补阳还五汤水提醇沉液为模型药物,拟以Hill量效方程为基础联合数学统计分析原理的谱效学数学模型,结合中药多成分指纹图谱和抗脑缺血症神经细胞损伤的药效学实验,计算补阳还五汤抗脑缺血症神经细胞损伤的多成分多靶点相对应的谱效学关系,为中药复方有效性研究提供新的思维、方法和实验依据。
     方法:重点是考察与评价补阳还五汤水提醇沉液抗脑缺血症神经细胞损伤及凋亡的谱效学关系。
     1.以Hill量效方程为基础,结合数学基本原理、统计学分析方法和计算机C语言程序等,初步建立中药复方谱效学数学模型。
     2.考察补阳还五汤水提醇沉液指纹图谱的HPLC测定条件以及该方中、盐酸川芎嗪、苦杏仁苷、芍药苷和阿魏酸的含量测定方法。
     3.考察补阳还五汤水提醇沉液在大鼠体内代谢的谱动学特征。
     4.采用溶度参数法获取补阳还五汤不同极性段有效成分群,并考察各成分群指纹图谱特征及相似度评价。
     5.通过星点设计考察与评价补阳还五汤水提醇沉液抗脑缺血症神经细胞损伤及凋亡的药效作用,结合先前建立的谱效学数学模型,计算补阳还五汤水提醇沉液抗脑缺血症神经细胞损伤和凋亡的谱效学参数。
     结果:1.建立了简便、准确、可靠的RP-HPLC法测定补阳还五汤水提醇沉液的指纹图谱。
     2.补阳还五汤水提醇沉液在SD大鼠体内代谢表观半衰期为9.108h,经9.108±1.96×9.062h后整方95%的成分被排除体外。
     3.采用11种不同溶度参数的溶剂可以分别提取分离得到补阳还五汤不同极性段成分群,且不同处理样品液指纹图谱相似度差异大,所含成分类别与含量也不同。
     4.结合谱效学数学模型、HPLC指纹图谱以及药效学实验,得到了补阳还五汤水提醇沉液抗脑缺血症神经细胞损伤的一系列谱效学参数,初步验证了先前建立的中药复方谱效学数学模型,且各成分对各靶点的作用系数差异大,表明各成分作用靶向与作用强度各有不同。
     结论:本实验所建立的中药复方谱效学数学模型得到了初步验证,能够应用于中药复方多成分多靶点特性的有效性研究,可以为中药现代化研究提供新的思路和实验依据。
The Buyanghuanwu decoction was a classical prescription for the treatment of the deficiency of Qi and blood stasis in traditional Chinese Medicine, which had a significant effect on cerebral ischemia, stroke, thrombosis, blood fat and improving immunity. This composition is numerous, complex and its validity basis and mechanism of action has not been elucidated. If we could build an effective research method adapted with tradition Chinese medicine compound characteristic, it was expected to provide new ideas and evidence for the Chinese medicine compound modernization study.
     Purpose:Take the Buyanghuanwu decoction after water extraction and alcohol precipitation as model drugs, this paper was intended to count the spectrum efficiency relationship of more ingredients corresponding more targets on resistance to nerve cells injury and apoptosis of brain ischemia symptomatic, which was used by its Chinese traditional medicine composition fingerprint and its pharmacodynamics experiment, bonded with the spectrum efficiency learn mathematics model, which based on Hill concentration-response equation combined with mathematical statistical analysis principle. It was hoped to provide new thinking, method and experimental basis to TCM compound effectiveness research.
     Methods:the key point is to examine and evaluate the spectrum efficiency relationship of more ingredients corresponding more targets on resistance to nerve cells injury and apoptosis of brain ischemia symptomatic.
     1. It established the TCM compound spectrum efficiency mathematics model initially based on Hill concentration-response equation combined with mathematics basic principle, statistical analysis method and the computer C language program.
     2. The fingerprint of Buyanghuanwu decoction after water extraction and alcohol precipitation was detected by HPLC, and the concentration determination method of ligustrazine, hydrochloric acid, amygdaline, herbaceous peony glucoside and ferulic acid was investigated by it.
     3. The spectrum metabolism characteristics of Buyanghuanwu decoction after water extraction and alcohol precipitation was investigated in vivo in SD rat.
     4. The different polarity composition groups in Buyanghuanwu decoction were obtained by the solubility parameter method and their fingerprint characteristics and similarity were evaluated.
     5. Through the star design for the investigation and evaluation of Buyanghuanwu decoction after water extraction and alcohol precipitation resistance to cerebral ischemia nerve cells injury and apoptosis, the spectrum efficiency relationship parameters were calculated by the previous spectrum efficiency mathematical model.
     Results:1. RP-HPLC methodology for fingerprint characteristics of Buyanghuanwu decoction after water extraction and alcohol precipitation was established with appropriate convenience and accuracy.
     2. The metabolism apparent half-life of Buyanghuanwu decoction after water extraction and alcohol precipitation was9.108h in rat in vivo, and it metabolized95%though9.108±1.96x9.062h after infusion.
     3. The different polarity composition groups for Buyanghuanwu decoction could obtain by11solvents processing with different solubility parameter, and the fingerprint similarity between different processing samples was remarkable differently, which showed different composition category and content.
     4. Combined with spectrum efficiency mathematical model, HPLC fingerprint and pharmacodynamics experiment, it has obtained a series of spectrum efficiency parameters of Buyanghuanwu decoction after water extraction and alcohol precipitation resistance to nerve cells injury and apoptosis of brain ischemia symptomatic. The spectrum efficiency mathematical model built before has been checked. In this experiment, spectrum efficiency parameters for every ingredient were of big difference, which showed that their targets and strength of the function were dissimilar.
     Conclusion:This study had validated the established spectrum efficiency mathematical model preliminary of Chinese herbal compound composition, which could be used in the Chinese herbal composition effectiveness research with many compound and many targets characteristics, and provide new ideas and experimental basis to the modernization of traditional Chinese medicine.
