机械推拿对慢性疲劳综合征脾虚型大鼠行为学和血清IL-1β及IL-6含量的影响

摘要
目的:利用自制的机械推拿器对慢性疲劳综合征脾虚型大鼠进行定时、定量的机械推拿治疗,观察其治疗疗效,从行为学和血清学角度探讨机械推拿对慢性疲劳综合征脾虚型大鼠的疗效和机制,进一步完善慢性疲劳综合征动物模型和为以后的推拿基础实验研究探索出一种更可靠的方法。
     方法:选取Wistar雄性大鼠108只(最后取72只统计),随机平均分为正常组、模型组和机械推拿组,每组再分为7天亚组、15天亚组,每亚组18只,采用应激法(单日冷水游泳,双日灌精炼猪油,每日食大白菜)建立慢性疲劳综合征脾虚证型大鼠模型,造模20天和综合行为学观察对大鼠模型进行评定。治疗组用机械推拿方式干预:机械推拿器在关元、足三里、天枢、脾俞、肝俞、太冲、三阴交(皆为双侧)推拿,每穴1min,力量120±20Gf;督脉、膀胱经(双侧)往返3次,频率120次/min,速度2.5mm/s,力量120±20Gf,每日一次。治疗15天。模型组只进行造模不治疗,正常对照组正常饲养不做处理。在治疗7天和15天后分别测定大鼠行为学变化,心脏穿刺采血,用ELISA法检测血清中白介素-1β,白介素-6含量的变化。所有数据均以(x±s)表示,应用统计软件进行统计处理,用One-Way ANOAY方差分析,均数间两两比较行SNK法Q检验,均以P<0.05或P<0.01是作为差异有统计学意义的标准。
     结果:(1)行为学观察方面,机械推拿组大鼠在机械推拿治疗后各项行为学观察指标好于模型组,有显著性差别,具有统计学意义,P<0.05或P<0.01;(2)对大鼠血清中白介素-1β,白介素-6检测发现,模型组比正常组升高,而治疗组与模型组相比较,白介素-1β,白介素-6的浓度降低,有统计学意义,P<0.05或P<0.01。
     结论:1.机械推拿可以提高慢性疲劳综合征脾虚型大鼠的体力及其神经系统兴奋性、增加其自发活动,能有效调节脾虚型慢性疲劳综合征大鼠
     2.机械推拿治疗慢性疲劳综合征大鼠的机制可能是通过降低血清中IL-1β, IL-6的含量,起到免疫调节作用的。
Objective:
     To observe the efficacy of treatment using a self-made mechanical massage device to treat rats with spleen deficiency type chronic fatigue syndrome (CFS) with a set time and set quantity of mechanical vibration massage therapy. From a behavior and serum analysis perspective, discuss the efficacy and mechanism of mechanical vibration massage on spleen deficiency type CFS to further improve the animal model of CFS and create a basis for future experimental research exploring massage as a reliable method of treatment.
     Methods:
     108 Wistar male rats (in the end,72 rats were used) were randomly divided into a normal group, a model group and a mechanical massage group. Each group was further divided into subgroups of 7 days and 15 days, with 18 rats in each subgroup. The CFS model was created using the stress method (cold water swimming on odd days, irrigation of refined lard on even days, and fed fresh cabbage daily). After 20 days of model construction, rat behavior was assessed to determine the success of the animal model. The treatment group used mechanical massage treatment:mechanical vibration at Guanyuan, Zusanli, Tianshu, Pishu, Ganshu, Taichong, Sanyinjiao (bilateral), every acupoint was treated for 1 minute using a strength of 120±20Gf; in addition, mechanical vibration was used on the Du meridian and the bladder meridian (bilateral) back and forth 3 times, with a frequency of 120 times/min, a speed of 2.5mm/s, and a strength of 120±20Gf, once a day. The model group underwent model construction but received no treatment, and the normal control group did not undergo model construction and received no treatment.7 days and 15 days after treatment, behavioral changes in the rats were measured and heart blood samples were taken. Using the ELISA method, serum interleukin-1βand IL-6 concentration changes were measured.
     Results:
     (1) Behavioral observation:The behavioral observation index of the mechanical massage treatment group was significantly better than that of the model group, P<0.05 or P<0.01. (2) Serum Interleukin-1β, Interleukin-6 concentrations:The model group had higher concentrations of serum Interleukin-1βand Interleukin-6 than the normal group, while the treatment group had lower concentrations of serum Interleukin-1βand Interleukin-6 than the model group, P<0.05 or P<0.01.
     Conclusion:
     1. Mechanical massage can improve physical strength in rats with spleen deficiency type CFS and can increase nervous system excitability, thereby increasing their spontaneous activity, and can effectively regulate rats with spleen deficiency type CFS.2. The mechanical massage mechanism of treatment for CFS may be related to decreased serum IL-1βand IL-6 levels, and play a role in immune regulation.
