BMP2mRNA在大鼠肝再生组织的表达与意义

摘要
目的:
     1、在基因水平上观察BMP2在肝组织的表达和肝再生过程中的动态变化特征。2、结合NF-κB在肝再生过程中的表达变化特征,探讨BMP2对肝再生的影响。
     方法:
     将健康、雄性Wistar大鼠随机分为正常对照组(NC)即0h组、假手术组(SO)和肝部分切除组(PH),采用原位杂交的方法测定NC组、SO组及PH组术后6h、l2h、24h、48h各时间点肝脏组织中BMP2mRNA的表达情况;采用免疫组化的方法测定以上各组肝脏组织中NF-κB的表达情况。
     结果:
     采用图像分析系统和统计学处理分析结果显示:1、NC组大鼠肝脏组织中有BMP2mRNA的表达。2、SO组与NC组大鼠肝脏组织中BMP2mRNA表达水平无明显差异。3、PH组术后6h,l2h,24h大鼠肝脏组织中BMP2mRNA表达水平与NC组及SO组相对应时间点比较存在统计学意义(P<0.05)。4、PH组术后6h,l2h,24h,48h各时间点组间BMP2mRNA表达率存在统计学意义(P<0.05)。5、PH组术后48h组与NC组BMP2mRNA表达率无明显统计学差异。6、NC组与SO组肝组织无NF-κB表达。7、PH组与NC组及SO组各时间点比较NF-κB表达率存在统计学意义(P<0.05)。8、PH组术后6h,l2h,24h,48h各时间点组间NF-κB表达率存在统计学意义(P<0.05)。9、BMP2mRNA与NF-κB的表达率相关性分析,表明BMP2与NF-κB在肝再生初期呈负相关(r =-0.860, P<0.05),在肝再生后期呈正相关(r =0.821, P<0.05)。
     结论:
     1、BMP2在正常大鼠肝组织中有表达且在肝再生过程中有变化。2、NF-κB在正常大鼠肝脏组织中无表达,在再生肝组织中有表达,且随肝再生时间进展其表达强度和表达部位有变化。3、BMP2可能抑制肝再生。
Objectives:1.at the level of genes, the expression of BMP2 in liver tissues and the characteristics of dynamic changes in the process of liver regeneration will be observed. 2. Combined with the characteristics of expression changes of NF-kB in the process of liver regeneration, the impact of BMP2 on the process will be explored.
     Methods: Healthy and male Wister rats were randomly divided into 3 groups—normal control group (NC) i.e. 0h group, sham-operated group (SO) and partial hepatectomy (PH). The expressions of BMP2mRNA in liver tissues of NC, SO and PH after 6h, 12h, 24h, 48h were observed using situ hybridization method; Immunohistoche-mistry method was adopted to determine the expressions of NF-kB in liver tissues of several groups said earlier.
     Results: Through the Image Analysis System and the Statistical Analysis, the following results can be shown: 1. the expression of BMP2mRNA exists in rat liver tissues of NC; 2. there is no significant difference in the expression levels of BMP2mRNA in rat liver tissues between SO group and NC group; 3. there exist statistical significance(p<0.05)comparing the expression of BMP2mRNA in rat liver tissues of PH after 6h, 12h, 24h, 48h with the BMP2mRNA expression of NC group and SO group in the corresponding moments; 4. there exists statistical significance (p<0.05) among the expression rats of PH groups after 6h, 12h, 24h, 48h; 5. there is no significant difference expression of BMP2mRNA of between PH group after 48h and NC group; 6. there is no NF-kB expression in liver tissues of NC group and SO group; 7. there exist statistical significance(p<0.05)comparing the expression of NF-kB in rat liver tissues of PH group and the NC group and SO group at different time points; 8. among the NF-kB expression rates of PH groups after 6h, 12h, 24h, 48h exists statistical significance (p<0.05); 9. through correlation analysis about expression rates of BMP2mRNA and NF-kB, such conclusion can be drawn that BMP2 and NF-kB are negatively correlated(r =-0.860, p<0.05)in the early period of liver regeneration and are positively correlated(r =0.821, p<0.05)in the latter period of liver regeneration.
     Conclusion: 1. there exists the expression of BMP2 in normal rat liver tissues and the change in the process of liver regeneration; 2. there is no NF-kB expression in normal rat liver tissues,NF-kB has the expression in the process of liver regeneration and changes in strength and position of its expression take place with the progress of liver regeneration’s time; 3. BMP2 may inhibits liver regeneration.
