刺五加提取物对仔猪抗氧化功能和肠道发育的影响

摘要
为了研究刺五加提取物对仔猪抗氧化功能和肠道发育的影响,本文选用胎次、体重相近的长白×大白二元杂交品种健康仔猪进行饲养试验。试验共用仔猪192头,随机分为8组,每组设3个重复,每个重复8头猪。空白对照组饲喂基础日粮,各试验组分别在基础日粮中添加50、100、200、1000mg/kg刺五加醇提取物,200mg/kg刺五加水提取物,10g/kg刺五加粗粉和200mg/kg金霉素。自仔猪出生7天起进行补饲,饲养试验期为5周,饲养试验结束后统计仔猪腹泻率与死亡率。
     本试验从每组分别随机选取14和21日龄仔猪3头,采集血清、肝脏和小肠组织。测定超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)的活性及丙二醛(MDA)的含量四个抗氧化指标和二糖酶、脂肪酶、胰蛋白酶三种消化酶的活性,并制作组织切片观察空肠与回肠的绒毛形态。
     对抗氧化功能的研究结果表明,50mg/kg刺五加醇提取物组与空白对照组相比,能够显著提高仔猪血清GSH-Px和肝脏SOD的活性;与200mg/kg金霉素组相比,能够显著提高仔猪血清GSH-Px和肝脏CAT的活性。200mg/kg刺五加水提取物组的仔猪血清MDA含量显著低于空白对照组。
     对肠道发育的研究结果表明,50mg/kg刺五加醇提取物组的仔猪空肠二糖酶和胰蛋白酶活性与空白对照组相比均有显著提高;与200mg/kg金霉素组相比,仔猪空肠二糖酶、脂肪酶和胰蛋白酶的活性均有显著提高。50mg/kg刺五加醇提取物组与200mg/kg刺五加水提取物组的仔猪空肠和回肠的绒毛发育良好,整齐细长;腹泻率与死亡率均为0,显著低于空白对照组和200mg/kg金霉素组。
     总之,50mg/kg刺五加醇提取物组和200mg/kg刺五加水提取物组的仔猪腹泻率和死亡率最低,为本试验的最佳添加剂量。
Studies in recent years found that the application of antibiotics in feed additives asgrowth promoters might induce bacterial resistance. Some countries have preventedantibiotics from using in feedstuff. Development and exploitation of new types of safe feedadditives to replace antibiotics were necessary. We chose Chinese herbal medicineAcanthopanax senticosus as feed additive in our research.
     In this paper, the effects of Acanthopanax senticosus extracts on antioxidative functionand intestinal development in piglets were studied. A total of 192 healthy Landrace×LargeYorkshire two-line crossbred piglets of seven-day old were randomly divided into eightgroups, each included three replicates of eight pigs. With the control of 0mg/kg to based diet,treatment groups were supplemented with 50, 100, 200, 1000mg/kg Acanthopanax senticosusethanolic extracts, 200mg/kg Acanthopanax senticosus aqueous extract, 10g/kg Acanthopanaxsenticosus powder, and 200mg/kg aureomycin, respectively. After seven and fourteen daysfeeding, three piglets were selected from each group randomly and Slaughtered to collectserum, liver, and intestine samples.
     The study on the antioxidative function indicated that the activities of glutathioneperoxidase (GSH-PX) in serum and superoxide dismutase (SOD) In liver of the pigletsincreased in the 50mg/kg Acanthopanax senticosus ethanolic extract treatment group incontrast to the control, and the activities of glutathione peroxidase (GSH-PX) in serum andcatalase (CAT) in liver of the piglets in this group increased in contrast to the the 200mg/kgaureomycin. The content of maleic dialdehyde (MDA) decreased in 200mg/kg Acanthopanaxsenticosus aqueous extract group compared with the control.
     For the intestinal development, there was increase at the activities of disaccharidase andtrypsin in the treatment group supplemented with 50mg/kg Acanthopanax senticosusethanolic extract compared with the control. And in contrast to the 200mg/kg aureomycingroup, the activities of disaccharidase, lipase and trypsin increased in this group. For thetreatment groups of 50mg/kg Acanthopanax senticosus ethanolic extract and 200mg/kgAcanthopanax senticosus aqueous extract, both of jejunal and ileal villus of piglets werehigher than the control, and the incidences of diarrhea and mortality in piglets were reduced.
     This study indicated that 50mg/kg Acanthopanax senticosus ethanolic extract and200mg/kg Acanthopanax senticosus aqueous extract were the optimal dose.
