通腑汤对急性肺损伤大鼠肺、肠组织CGRP的影响

摘要
目的:观察通腑汤对急性肺损伤大鼠肺与肠组织CGRP水平的影响,探讨通腑汤治疗急性肺损伤的作用机制,从而为通腑汤在临床上的应用提供科学的理论依据。
     方法:选取健康Wistar大鼠180只,随机分为6组。即正常对照组、模型组、地塞米松组、中药低剂量组、中药中剂量组、中药高剂量组、30只/组。尾静脉注射LPS复制ALI模型,半小时后中药各剂量组灌胃、地塞米松组腹腔注射治疗,于治疗后第1、2、6小时,每组每次各10只,处死大鼠。光镜下观察肺、肠的组织形态学变化;ELISA法同步检测肺、肠组织中CGRP的含量。
     结果:
     1、肉眼见模型组肺、肠组织颜色暗红、肿胀、出血、各治疗组较模型组减轻。
     2、光镜下见模型组肺、肠组织炎性细胞浸润、水肿、充血、结构破坏,各治疗组较模型组程度减轻。
     3、模型组肺、肠组织CGRP含量在前6小时内随时间的延长而增长。
     4、ALI肺、肠组织中:模型组CGRP含量高于正常对照组,有统计学意义(P<0.05);地塞米松组、中药各剂量组CGRP含量均低于模型组,有统计学意义(P<0.05);中药低剂量组CGRP含量高于地塞米松组,有统计学意义(P<0.05);中药中剂量组、中药高剂量组CGRP含量与地塞米松组无差异,无统计学意义(P>0.05);中药中剂量组、中药高剂量组CGRP含量均低于中药低剂量组,有统计学意义(P<0.05);中药高剂量组CGRP含量与中药中剂量组无差异,无统计学意义(P>0.05)。
     结论:
     1、通腑汤可降低ALI大鼠肺组织中CGRP的含量,高剂量组与中剂量组疗效较好。
     2、通腑汤可降低ALI大鼠肠组织中CGRP的含量,高剂量组与中剂量组疗效较好。
Objective:To observe the effect on CGRP in ALI rat model lung and intestine, and to approach the mechanism of Tongfu Decoction anti-ALI, so to provide experimental base on the lung and the large intestine being interior-exteriorly related theory.
     Method:180 healthy wistar rats were choosed and randomly divided into six groups:the normal control group, the model group, hexadecadrol group, Chinese herbal medicine low-dose group, medium-dose group, and high-dose group,30 rats each group. Copying ALI model by caudal vein injecting LPS,giving respectively the Chinese herbal medicine intragastric administration and Dexamethasone intraperitoneal injection half an hour later,and executing rats in 1、2、6 hour after treatment respectively.10 rats each time in each group. We observed lung and intestine histomorphology changes, detected CGRP contene in lung and intestine by ELISA method synchronously.
     Result:
     1 Lung and intestine tissue in the model group showed dark red, turgent, bleeding by naked eye, but other treatment groups showed lighter than it.
     2 Lung and intestine tissue in the model group showed inflammatory cell infiltrate, edema, hyperemia, disorganization in light microscope, but other treatment groups showed lighter.
     3 CGRP content increased as time extending in first 6 hours in lung and intestine tissue of the model group.
     4 Lung and intestine tissue in ALI:CGRP contents in the model group were higher than the normal control group (P<0.05); contents in hexadecadrol group and Chinese herbal medicine groups were lower than the model group (P<0.05); contents in Chinese herbal medicine low-dose group were higher than hexadecadrol group (P<0.05); contents in Chinese herbal medicine medium-dose group and high-dose group showed lower than low-dose group (P<0.05); Chinese herbal medicine medium-dose group and high-dose group had no statistical significance (p>0.05).
     Conclusions:
     1 Tongfu Decoction could decrease CGRP in lung tissue of ALI rats model, The curative effects of high-dose and medium-dose group were better.
     2 Tongfu Decoction could decrease CGRP in intestine tissue of ALI rats model, The curative effects of high-dose and medium-dose group were better.
