补脾益肠汤对溃疡性结肠炎模型小鼠血清白介素8水平影响的研究

摘要
[目的]:探讨补脾益肠汤治疗UC的作用机理,本研究选用与临床溃疡性结肠炎病理特点相似的实验性溃疡性结肠炎动物模型,观察药物的作用,并从炎症因子(IL-8)方面探讨其疗效机理,为中医药治疗UC提供依据。
     [方法]:将100只昆明小鼠,随机分为5组,分别为正常组、模型组、中药高剂量组、中药低剂量组、西药组。除正常组外,其余小鼠均以葡聚糖硫酸钠(DSS)自由饮用复制溃疡性结肠炎模型。每日观察小鼠的体重变化、大便性状和隐血情况。在急性期和慢性期分别取小鼠结肠组织作病理,评价组织学损伤程度。用放射免疫法检测各组小鼠血清中的IL-8含量,分析补脾益肠汤对IL-8的影响。
     [结论]:给予药物治疗后,UC小鼠血清中IL-8明显降低,光镜下病理组织炎细胞浸润程度也明显降低,炎症症状得到明显改善,且与西药治疗效果相比无显著性差异,这说明补脾益肠汤对葡聚糖硫酸钠所致小鼠慢性溃疡性结肠炎动物模型有治疗作用。补脾益肠汤治疗UC机理之一与其抑制促炎因子IL-8有关。
[Aim]:In order to explain the effect of BPYCT, we investigated the effect of it on treating experimental UC in mice.We also study the connection between IL-8 and UC to explain the effect of the recipe.
     [Methods]:We devide 100 mice into 5teams, including normal, model, high Chinese herb, low Chinese herb and SASP.Give mice DSS to make UC models and observe the weight, stool and hiden blood of mice.Choose the colon-tissue of mice to evaluate the damage-level.At last, measure the IL-8 amount of blood and analyse what can the recipe do to IL-8.
     [Results]:After therapy, IL-6 level decreases.The pathologic changes of colon mucosa and symptoms are both improved.This experiment shows that BPYCT is effective to DSS models.This recipe can improve the symptoms.The mechanism may be related to inhibiting inflammatory factors (IL-8).
