芒果甙对血小板4因子,血浆β球蛋白释放以及血小板聚集率的影响

摘要
目的:通过体外实验,研究芒果甙对血小板释放血小板4因子(PF4)和血浆β球蛋白(β-TG)的影响。以及芒果甙对血小板聚集率的影响。并初步探讨芒果甙影响血小板聚集和释放功能的作用机制。
     方法:收集健康志愿者的外周全血约10ml,再加入待测不同浓度的芒果甙以及阿司匹林,用胶原作为诱导剂,采用全血电阻法Chrono-Log血小板聚集分析仪测定血小板聚集率,同样方法再抽取4ml全血,用ELISA方法,检测外周血血浆中PF4、β-TG的含量。
     结果:1 .ELISA实验法检测血小板4因子和血浆β球蛋白:阿司匹林组β-TG含量增加(P<0.01),3组不同浓度组芒果甙β-TG也显著增加(P<0.05),阿司匹林组PF-4含量增加(P<0.01),3组不同浓度组芒果甙PF-4也含量增加(P<0.01),低浓度组和中高两浓度组之间有显著差异(P<0.01),但后两者之间差异不明显。
     2.全血电阻法Chrono-Log血小板聚集分析仪测定血小板聚集率:正常对照组与低浓度芒果甙组间无明显差异,P值为0.999,但与中浓度组,高浓度芒果甙组间有显著差异,P值分别为0.046以及0,均<0.05。正常对照组与阿司匹林组间也有明显差异,P值为0.024。低浓度芒果甙组与中浓度组间并无统计学意义,P值0.244.>0.05,但与高浓度芒果甙间有统计学意义,P为0。中浓度芒果甙组与高浓度组间P值0.024,<0.05,具有统计学意义。阿司匹林实验组只与高浓度芒果甙组间有统计学差异,P值0.046。
     结论:芒果甙可以抑制血小板的释放,呈浓度依赖性(随着浓度增高,呈递增趋势),但不能降低血小板释放PF4和β-TG的表达率。同时,芒果甙也可以抑制血小板的聚集,并呈浓度依赖性。
Objective: In vitro experiments, to study the platelet release of mangiferin on platelet factor 4 (PF4) andβ-TG .And the mangiferin effect on platelet aggregation. To investigate the mangiferin function of platelet aggregation and the emancipation mechanism.
     Methods: Collection of peripheral blood from healthy volunteers about 10ml, and then adding different concentrations of mangiferin and aspirin, with collagen as inducing agents, then use the Chrono-Log to measure platelet aggregation .By the same Method, collected 10ml of whole blood, with the ELISA method to detect in plasma PF4,β-TG content.
     Results:1. ELISA test method of platelet factor 4 and plasmaβglobulin: Forβ-TG ,the group of aspirin and 3 teams of mangiferin are increased significantly (P<0.05). For PF-4,the group of aspirin and 3 teams of mangiferin are increased significantly (P<0.01). But the low concentration group and the two high concentration have obvious differences (P<0.01).
     2. Resistance method of whole blood to analyzer by the Chrono-Log platelet aggregation: Normal control group and the low concentration of mangiferin was no significant difference between groups, P value of 0.999, but the concentrations, high concentrations of mangiferin were significantly different between groups, P values were 0.046 and 0, all <0.05. The normal control group and aspirin are also significant differences between groups, P value of 0.024. Low concentration in the concentration of mangiferin group and between groups was not statistically significant, P = 0.244> 0.05, but with high concentrations of mangiferin were statistically significant, P =0. Concentration and high concentration of mangiferin group between groups, P = 0.024 <0.05, with statistical significance. Aspirin experimental group and high concentrations of mangiferin significant difference between groups, P = 0.046.
     Conclusion: Mangiferin can inhibit platelet emancipation ,and by concentration trend (as the concentration increased, showed a decreasing trend).But the expression of platelet release PF-4 andβ-TG can not be reduced. Meanwhile, mangiferin also inhibits platelet aggregation in a concentration dependent manner.
