ABCG2在肾癌中的表达及其与肾癌干细胞关系的初步研究

摘要
目的探讨三磷酸腺苷结合转运蛋白G超家族成员2(ABCG2)在肾癌组织中的表达及其与肾癌干细胞之间的关系。
     方法采用免疫组化方法检测65例手术切除肾癌组织(透明细胞癌)和15例癌旁正常组织中ABCG2的表达情况。应用RT-PCR方法检测30例肾癌患者的癌组织及10例正常肾组织中ABCG2的mRNA表达。
     结果ABCG2阳性染色主要定位于癌细胞细胞膜,胞质中也有部分表达。ABCG2在肾癌组织中呈较高表达,其中ABCG2在低分化肾癌组织中的表达高于中、高分化者(P<0.05);ABCG2的表达与肾癌患者的临床分期无关(P>0.05)。癌旁正常组织中ABCG2呈弱表达或无表达,与肾癌组织相比差异性显著。肾癌组织中ABCG2 mRNA水平的表达明显高于肾癌旁正常组织(P<0.05),与免疫组化结果一致。
     结论ABCG2在肾癌组织中表达较高,其可能参与了肾癌的发生和发展,可以作为肾癌干细胞研究的重要标志物。
[Objective] To investigate the expression and significance of ATP-binding cassette transporter (ABCG2) in renal carcinoma tissues and the relationship between the ABCG2 and renal carcinoma stem cell.
     [Methods] Expression of ABCG2 protein was examined by immunohistochemical technique in specimens from 65 renal carcinoma tissues( clear cell carcinoma) and 15 surronding normal tissues. The expression of ABCG2 mRNA was measured by RT-PCR in specimens from 30 renal carcinoma tissues and 10 surrounding normal tissues.
     [Results] The study confirmed the predominant localization of ABCG2 transporter in plasma membrane of the carcinoma cell , only little part was localization in cytoplasma of the tumor cell. ABCG2 protein expression was high in the renal carcinoma cell, while in normal renal tissues the expression was low or no expression. The positive rates of ABCG2 expression in Undifferentiated carcinoma was significantly higher than those in well and/or moderately differentiated carcinoma (P<0.05). The mRNA expression of ABCG2 was significantly higher in renal carcinoma tissue than in normal renal tissues(P<0.05).
     [Conclusion] The expression of ABCG2 in renal carcinoma tissue was high. ABCG2 may be closely related to the oncogenesis and progress of renal cancer, and may be an important factor in the research of renal cancer stem cell.
