榆黄蘑的化学成分研究

摘要
榆黄蘑又名金顶蘑或称金顶侧耳(pleurotus citrinopileatus sing.),属于担子菌亚门,层菌纲,伞菌目,侧耳科,侧耳属,是一种珍贵的经济菌物。人们常食用,味美,香甜,具有一定的医疗保健作用。近年来研究表明榆黄蘑主要含有甾醇类和脂肪酸等成分。但目前对榆黄蘑有效成分的研究并不全面,药理学的研究也刚刚起步。作者通过硅胶柱层析,凝胶柱层析等分离手段从榆黄蘑中分离出8个单体化合物,通过理化性质和现代波谱学方法和技术(ESI-MS、~1HNMR、~(13)CNMR等)分析,确定他们的结构分别为:甘露醇,麦角甾醇,3β,5α,6β-三羟麦角甾醇,腺嘌呤核苷,尿嘧啶,烟酰胺,肌苷,鸟苷。
     利用HPLC—MS/MS对榆黄蘑中甘露醇进行定量。用MS/MS作为HPLC的检测器采用多反应检测(MRM)扫描方式,建立了HPLC-MS/MS测定榆黄蘑中甘露醇含量的方法。该方法的线性范围为0.208-1.04mg·mL~(-1)。通过样品的加标试验,回收率为97.12%,精密度RSD(n=5)为0.94%。实际样品分析表明,榆黄蘑中甘露醇的含量为38.9mg/g。本方法选择性高、灵敏、快速。也适用于其他复杂体系中甘露醇含量的快速测定。
Pleurotus citrinopileatus is belong to Basidiomycotina Hymenomycetes class Agaricales Pleurotus is one kind of precious economical fungus,People often edible, the taste is beautiful,fragrant,has certain healthcare function.This year studies indicated that Pleurotus citrinopileatus mainly includes fatty acid ingredients and so on sterol class.But present is not comprehensive to the Pleurotus citrinopileatus effective component's research.The Pharmacology research also just started.The author through the silica gel column chromatographic analysis,separation methods and so on gelatin column chromatographic analysis separates the monomer compound from Pleurotus citrinopileatus,Through physical and chemical properties,spectrum of modern methods and techniques(ESI-MS,1HNMR,13CNMR),Determined that their structure respectively is mannitol,ergosterol,ergotsa-7,22-dien-3β,5α,6β-triol,adenosine,uracil, nicotinamide,inosine,guanosine.
     The mannitol carries on the quota using HPLC-MS/MS to Pleurotus citrinopileatus.Uses the multi-response examination with MS/MS as the HPLC detector (MRM) to scan the way,Has established in HPLC-MS/MS determination Pleurotus citrinopileatus the mannitol content method.This method's linearity range is 0.208-1.04mgmL-1.Through the sample Canadian sign experiment,the returns-ratio is 97.12%,accuracy RSD(n=5) is 0.94%.The actual sample analysis indicated that in Pleurotus citrinopileatus the mannitol content is 38.9mg/g.This method selectivity high, is keen,is fast.It also suitable in other complex systems the mannitol content fast determination.
