树突状细胞在变应性鼻炎小鼠发病中的作用研究

摘要
目的
     探讨树突状细胞(dendritic cells, DC)在变应性鼻炎(allergic rhinitis, AR)中的作用,以及AR和支气管哮喘之间的联系。
     方法
     6-8周BALB/C小鼠采用卵清白蛋白致敏激发建立AR小鼠模型;免疫荧光双标组织化学染色检测AR小鼠和正常对照组小鼠鼻粘膜中DC的分布及形态;取C57B/L小鼠骨髓细胞体外分离培养未成熟DC,Hoechst33342荧光标记后,注射入小鼠鼻粘膜下,追踪注入的DC在两组小鼠鼻粘膜及肺脏中的分布,采用免疫荧光双标组织化学染色方法观察其表型变化。
     结果
     与正常对照组小鼠相比,AR组小鼠打喷嚏和挠鼻次数明显增多,鼻粘膜上皮纤毛紊乱脱落,固有层内可见嗜酸性粒细胞、炎性细胞浸润。AR组小鼠DC主要分布于鼻粘膜上皮层和固有层,而且主要处于成熟状态。正常对照组小鼠鼻粘膜内未见或罕见DC分布。于两组小鼠鼻粘膜下注入Hoechst33342荧光标记未成熟DC,AR组注入的DC分布于小鼠鼻粘膜固有层和上皮层,而且转化为成熟状态,在一例AR小鼠的肺脏切片中发现有注入的DC,主要分布于支气管粘膜旁及肺间质内;正常对照组注入的DC主要分布于小鼠鼻粘膜上皮层,主要保持未成熟状态,小鼠的肺脏切片中未发现有注入的DC。
     结论
     DC可能在小鼠变应性鼻炎的发病以及变应性鼻炎与支气管哮喘的联系中发挥重要作用。
Objective
     To study the role of dendritic cell (DC) in allergic rhinitis and the relationship between allergic rhinitis and allergic asthma.
     Methods
     BALB/C mice, 6~8 weeks old, were randomly divided into two groups: allergic rhinitis (AR) group and control group. AR model in mice were established by ovalbumin(OVA) intraperitoneal immunization and nasal antigen challenge. Behavior score and the routinely HE staining of the nasal mucosa were used to evaluate the AR model. DC numbers were evaluated by immunohistochemistry. The mice in the AR group and the control group, received an intranasal injection of immature DC obtained from the bone marrow and marked with Hoechst33342. Then we traced the immature DC by immunofluorescence histochemistry to observe the changes of the injected DC.
     Results
     The behaviors of the AR mice were different from the mice of control group. Increased numbers of DC can be found in mice with AR. The immature DCs which were injected intranasaly distributed in the lamina propria and epithelium of the mucosa in AR mice and turned to be mature ones . In one of the AR mice the injected DCs transferred to the lung. While in the control group , the injected DCs stayed to be immature ones in the epithelium of the mucosa. No injected DCs were found in the lung of the mice in the control group.
     Conclusions
     DC may play an important role in allergic mice. And the allergic rhinitis maybe induce asthma by the migration of DC.
