OT和Ghrelin在小鼠下丘脑的分布及其与肥胖的关系

摘要
为了探讨OT和Ghrelin在小鼠下丘脑的分布及其与肥胖的关系,本研究采用免疫组化SABC法对OT和Ghrelin在正常小鼠和肥胖小鼠下丘脑调节摄食的核团中表达变化进行了研究,并研究了注射外源性催产素对小鼠下丘脑中Ghrelin表达量的变化,其结果可为动物能量代谢、神经肽之间相互调控关系和肥胖的机理研究提供重要资料。
     主要结果如下:
     1.催产素在小鼠下丘脑弓状核、腹内侧核、室旁核(正常小鼠)中分布较多,而在背内侧核、视交叉上核、下丘脑外侧区中分布较少;Ghrelin在弓状核、下丘脑外侧区、背内侧核、室旁核(肥胖小鼠)中分布较多,而在视交叉上核、腹内侧核中分布较少。表明催产素和Ghrelin在不同核团表达量的差异和所在核团的摄食调节功能相吻合。
     2.与正常小鼠相比,肥胖小鼠下丘脑室旁核中催产素的表达量显著减少(P<0.01)。而在下丘脑其他摄食核团中催产素的表达量与正常小鼠基本相同,且不存在性别差异。
     3.与正常小鼠相比,肥胖小鼠下丘脑室旁核、背内侧核和下丘脑外侧区中Ghrelin的表达量显著增多(P<0.01)。
     4.给肥胖小鼠连续4周腹腔注射催产素后,下丘脑背内侧核、室旁核及下丘脑外侧区Ghrelin的表达量显著下降,在弓状核、腹内侧核及视交叉上核Ghrelin表达量也有一定的下降;雄性肥胖小鼠下丘脑各核团中Ghrelin的表达量的下降幅度比雌性肥胖小鼠大一些。
     结论:催产素能使下丘脑室旁核、背内侧核、下丘脑外侧区、弓状核、腹内侧核及视交叉上核中Ghrelin的表达量下降,催产素和Ghrelin在中枢摄食调节中存在着相互拮抗的关系。提示这是催产素减肥作用的途径之一。
To study the distuibution of oxytocin(OT) and Ghrelin in hypothalamus and its relationship with obesity, so immunohistochemical SABC method was employed to determine the expression of OT and Ghrelin in the hypothalamic nuclei controlling food intake of normal mice and obese mice.And the effects of exogenous oxytocin (OT) on Ghrelin expression on hypothalamus in obese mice were studied in intraperitoneal(ip) injection by immunohistochemical SABC method. The result can play an important role in revealed energy metabolism, the interregulate of neuropeptide,and the treatment of obesity.The main result as follows:
     1. The distribution of oxytocin in the ARC、VMN and PVN(normal mice) are more,but in the DMN、SChN and LHA are less; The distribution of Ghrelin in the ARC、LHA、DMN and PVN(obese mice) are more, but in the SChN、VMN are less.The results suggested the differences of oxytocin (OT) and Ghrelin expression in different nuclear unites are identical with the regulating function of food intake in the nuclear unites.
     2. Compared to normal mice, expression of OT in PVN of obese mice was decreased notably(P<0.01). But it is identical of the expression in the other nuclear unites, and no change of sex difference was found.
     3. Compared to normal mice, the expression of Ghrelin was increased notably in PVN、DMN and LHA in obese mice(P<0.01).
     4. After intraperitoneal(ip) injection of OT for 4 weeks to obese mice, the expression of Ghrelin was decreased notably in PVN、DMN and LHA,and it was also decreased in the ARC、VMN、SChN slightly. The decrease amplitude of the Ghrelin of male is more than female.
     From the results above, we draw the following conclusions: OT inhibited the expression of Ghrelin in PVN、DMN、LHA、ARC、VMN and SChN, OT has the mutually antagonistic relationship with Ghrelin. This prompted OT has the antiobesity action, reducing the expression of Ghrelin in the regulation of food intake in the central nervous system is one of way of its role.
