CD147在亲环素A诱导Jurkat细胞增殖、活化和趋化过程中的作用

摘要
目的
     研究CD147分子在亲环素A(CyPA)诱导的人T系白血病细胞株Jurkat细胞增殖、活化和趋化过程中的作用,旨在明确CyPA在Jurkat细胞中可能的作用及机制,并探讨CyPA和CD147分子的相互作用对肿瘤细胞生物学行为的影响。
     方法
     1.采用RT-PCR法检测转染CD147siRNA的Jurkat细胞(已构建)CD147 mRNA水平,证实转染的有效性;
     2.以不同浓度CyPA(0.01nM,0.1nM,1nM,10nM)处理Jurkat细胞24 h和48 h,采用四甲基偶氮唑盐(MTT)法检测CyPA的促细胞增殖作用;以最佳CyPA作用浓度处理Jurkat、Jurkat-vector和Jurkat-CD147siRNA细胞,检测CD147siRNA转染对CyPA促细胞增殖的影响;
     3.以10nM CyPA处理Jurkat、Jurkat-vector和Jurkat-CD147siRNA细胞24 h,采用流式细胞仪法检测CyPA处理前后细胞表面活化标记分子CD25的表达;
     4.采用Transwell小室法检测CyPA趋化Jurkat、Jurkat-vector和Jurkat-CD147siRNA细胞的能力;
     结果
     1.CD147siRNA转染有效,Jurkat-CD147siRNA细胞的CD147mRNA表达水平较Jurkat细胞显著下降,抑制率为67.12%(P<0.05);
     2.CyPA以浓度依赖性方式促进Jurkat细胞增殖,CyPA浓度为0.1nM时开始促细胞增殖,10nM时促增殖作用最强;CD147siRNA转染导致CyPA促细胞增殖作用显著下降,以Jurkat+CyPA组为对照,Jurkat-CD147siRNA+CyPA组在24 h和48 h时的增殖抑制率分别为38%和27%,差异有统计学意义(P均<0.05);
     3.CyPA可促进Jurkat细胞的活化,CyPA处理前后的CD25阳性表达率分别为6.28%和7.75%(P<0.05);CD147siRNA转染致CyPA促细胞活化能力下降,Jurkat-CD147siRNA+CyPA组和Jurkat+CyPA组的CD25阳性表达率分别为3.51%和7.75%,有显著性差异(P<0.05);
     4.CyPA对Jurkat细胞有趋化作用,CyPA趋化组与对照组趋化细胞数分别为67611±4148和29056±3643,差异有统计学意义(P<0.05);CyPA趋化Jurkat-CD147siRNA的细胞数目较趋化Jurkat细胞显著减少(P<0.05)。
     结论
     1.CyPA促进Jurkat细胞增殖、活化和趋化。
     2.CD147分子在CyPA诱导的Jurkat细胞生物学特性改变过程中发挥作用。
Objective
     To investigate the roles of CD147 on the proliferation,activation and chemotaxis of Jurkat cell induced by Cyclophilin A(CyPA).To identify the possible function and mechanism of CyPA on Jurkat cells and to investigate the role of interaction of CyPA and CD147 on tumor progression.
     Methods
     1.CD147 mRNA level of CD147siRNA transfected Jurkat cell was identified by semi-quantitive reverse transcription-polymerase chain reaction(RT-PCR)to assess the validity of tranfection;
     2.Jurkat cells were treated with different concentrations of CyPA (0.01 nM,0.1 nM,1 nM,10 nM)for 24 h and 48 h.MTT colorimetric assay was used to detect the proliferation of Jurkat cell induced by CyPA; Jurkat,Jurkat-vector and Jurkat-CD147siRNA cells were treated with optimal CyPA concentration,CD147siRNA transfection on the proliferation activity of Jurkat induced by CyPA were detected;
     3.Jurkat,Jurkat-vector and Jurkat-CD147siRNA were treated with 10nM CyPA for 24 h,Flow cytometry(FCM)was used to analyse CD25 expression of each cell group;
     4.Transwell chamber was used to detect the chemotactic ability of CyPA for Jurkat,Jurkat-vector and Jurkat-CD147siRNA cells;
     Results
     1.Compared to Jurkat cell,CD147 mRNA of Jurkat-CD147siRNA cell decreased significantly by 67.12%(P<0.05),which proved the validity of transfection;
     2.CyPA stimulated the proliferation of Jurkat cells in a dose-dependent manner.CyPA began to promote proliferation at concentration of 0.1 nM,the effect reached the peak at l0 nM. Transfection of CD147siRNA resulted in decreased cell proliferation induced by CyPA.Compared to Jurkat+CyPA group, Jurkat-CD147siRNA +CyPA group exhibited proliferation inhibition rate of 38%at 24 h and 27%at 48 h(P<0.05);
     3.CyPA promoted the activation of Jurkat cell.The percentage of CD25 positive cells of CyPA-untreated and CyPA-treated Jurkat cells were 6.28%and 7.75%,respectively(P<0.05);transfection of CD147siRNA decreased the cell activation rate induced by CypA.The percentage of CD25 positive cell of Jurkat-CD147siRNA+CyPA group and Jurkat+CyPA group were 3.51%and 7.72%,respectively(P<0.05).
     4.CyPA exhibited chemotactic activity for Jurkat cell.Compared to control group,the number of cells migrating to CyPA-containing chemotactic solution and to control chemotactic solution was 67611±4148 and 29056±3643,respectively(P<0.05);the cell number of Jurkat-CD147siRNA migrating to CyPA decreased significantly than Jurkat cell(P<0.05).
     Conclusion
     1.CyPA promote proliferation,activation and chemotaxis of Jurkat cell;
     2.CD147 play roles in the CyPA-induced biological function of Jurkat cell.
