摘要
本文通过对不同发育阶段白毛黑眼(WHBE)兔外周血和骨髓红细胞免疫学的变化研究,结合红细胞膜结构和功能、红细胞免疫调控淋巴细胞、自然杀伤(NK)细胞活性与粘附肿瘤细胞能力等指标,探讨红细胞免疫与衰老、肿瘤的关系,并与日本大耳白兔比较,揭示WHBE兔红细胞功能的潜在特性,为WHBE兔在生物医学研究领域中的应用提供实验依据。
     研究结果:
     (1) WHBE兔外周血和骨髓红细胞C3b受体花环率(E-C3bRR)、红细胞免疫粘附促进因子(RFER)与年龄呈显著负相关,而红细胞免疫复合物花环率(E-ICR)、循环免疫复合物(CIC)和红细胞免疫粘附抑制因子(RFIR)与年龄呈显著正相关;且老年WHBE兔骨髓E-C3bRR和RFER活性明显高于日本大耳白兔。说明机体衰老引起WHBE兔红细胞免疫功能增龄性降低,且WHBE兔红细胞免疫功能略高于日本大耳白兔。
     (2) WHBE兔红细胞膜超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、Na+-K+-ATPase、Ca2+-Mg2+-ATPase、谷草转氨酶(GOT)、唾液酸酶(SA)、胆固醇(CHO)、乳酸(Lac)、高密度脂蛋白(HDL-C)和低密度脂蛋白(LDL-C)与年龄和红细胞免疫功能密切相关,日本大耳白兔的变化规律亦类似,说明WHBE兔衰老过程中体内自由基生成增多,抗氧化能力下降以及糖、脂代谢的紊乱,导致红细胞结构和形态的损害,影响红细胞功能的发挥,最终引起机体红细胞免疫功能的降低。
     (3)青年WHBE兔红细胞促淋巴细胞增殖能力(SI)明显高于幼年和老年组;WHBE兔红细胞促NK细胞活性随增龄性升高,并与红细胞免疫功能显著相关,且老年雌性WHBE兔红细胞促NK细胞活性的能力明显高于日本大耳白兔,说明红细胞参与调控淋巴细胞和NK细胞活性,其中青年动物的红细胞能更好地促进淋巴细胞的免疫调控,而老年动物的免疫活性细胞较多地处于预激活状态,对刺激显示良好的免疫应答能力。
     (4) WHBE兔红细胞免疫粘附肿瘤细胞的能力(EIAT)随增龄而降低,并与CIC和红细胞免疫功能显著相关,且老年WHBE兔EIAT显著高于日本大耳白兔,说明WHBE兔红细胞具有粘附、识别和杀伤肿瘤细胞的作用,且与红细胞免疫功能的降低和CIC的升高密切相关。
     以上研究结果表明,WHBE兔和日本大耳白兔红细胞免疫功能既有随增龄性变化规律的共性,又有其品种间的差异,且WHBE兔红细胞免疫功能略高于日本大耳白兔,表明WHBE兔具有独特的红细胞生物学特性,为WHBE兔在生物医学研究领域中的应用尤其以红细胞为对象的研究中提供良好的实验材料和生物学参考数据,同时为人类衰老和红细胞免疫低下的研究奠定理论基础。
The study focused on the peripheral blood and bone marrow erythrocytic immunofunctions in WHBE (white hair and black eyes) rabbits and Japanese white rabbits. To discuss the relation between erythrocytic immunofunctions, age and tumor, and further to explore potential characteristics of WHBE rabbits, it will provide an experiment basis for biomedical applications of erythrocytic immunofunctions in WHBE rabbits, we researched the changes of the structure and function of erythrocyte membrane, erythrocyte immunoregulation of lymphocyte, natural killer (NK) cell activity, and the ability of erythrocyte adhering to tumor cell (EIAT).
     The results as follows:
     1. Negative correlation between age and E-C3bRR, RFER, while significantly positive correlation was found between age and E-ICR, CIC, RFIR in WHBE rabbits.The bone marrow E-C3bRR and erythrocyte immune adherence enhancing factor in senile WHBE rabbits were obviously higher than those in Japanese white rabbits, which indicated that the erythrocytic immunofunctions decreased with advancing age, and caused by body senescence. The erythrocytic immunofunctions of WHBE rabbits were slightly stronger than those of Japanese white rabbits.
     2. Closely correlation between the activities of erythrocyte membrane SOD, GSH-px, MDA, Na+-K+-ATPase, Ca2+-Mg2+-ATPase, GOT, serum SA, serum CHO, serum Lac, serum HDL-C, serum LDL-C, and age, erythrocytic immunofuctions, and these changes were similar to Japanese white rabbits, which indicated that the production of free radical increased in both WHBE rabbits and Japanese white rabbits body with ageing process, resulting in the reduced capacity of oxidation resistance and the disordered glucose metabolism and lipid metabolism, and erythrocyte was damaged and influencing the function of erythrocyte, and caused the erythrocytic immunofunctions reducing.The erythrocyte membrane function of WHBE rabbits were slightly stronger than those of Japanese white rabbits.
     3. The index of lymphocyte proliferation in youth group was significantly higher than that in juvenile group and senile group, and the activity of NK cell in WHBE rabbits increased with advancing age, and these changes correlated with erythrocytic immunofunctions.The NK cell proliferation in female WHBE rabbits was obviously higher than that in Japanese white rabbits.Those indicated that erythrocyte can participate to regulate lymphocyte and NK cell activities, and the erythrocyte in the youth period can increase the lymphocytic regulatory ability to reaching the best status. Furthermore, most of the immune competent cells in senile group were in pre-activated status and displayed a well immune response by activated.
     4. The ability of erythrocyte adhering to tumor cell in WHBE rabbits decreased with advancing age, and these changes were siginificant correlated with CIC and erythrocytic immunofunctions, furthermore, those of senile WHBE rabbits were obviously higher than those of Japanese white rabbits, which indicated that erythrocyte in WHBE rabbits have adhesion, identification and kill tumor cells effects, and it closely related with lowing erythrocyte immunofunction and increased CIC.
     In conclusion, the reythrocytic immunofunction in WHBE rabbits and Japanese white rabbits not only have mutual change patterns of rabbit strains with advancing age, but also have difference between strains, and erythrocytic immonfunctions in WHBE rabbits were slightly higher than those in Japanese white rabbits, and those were revealed the erythrocyte biological function characteristics in WHBE rabbits and plentied its biological characteristics. It will provide a better experiment animal and abundant biological data for the future application in biomedical research and erythrocytic immunofunction studies, and establish rational for human ageing and deficient of erythrocytic immuofunction.
