吸烟及戒烟对大鼠血清瘦素、脂联素等炎症因子水平的影响

摘要
目的通过研究不同吸烟量及戒烟对大鼠血清瘦素、脂联素、白细胞介素(interleukinIL)-6及C反应蛋白(C-reactive protein CRP)水平的影响,探讨瘦素、脂联素等炎症因子在吸烟所致COPD全身炎症反应中的作用。
     方法8周龄健康雄性Wistar大鼠(山西医科大学实验动物中心提供)32只,体重(200±20)g,随机分为健康对照组、大量吸烟组、小量吸烟组、大量吸烟后戒烟组,每组各8只。参照许三林等自制实验性大鼠被动吸烟装置,大量组每天吸烟2次(上、下午各一次),每次吸烟10支,大约两个小时,每周吸烟6天,共吸烟20周。小量组每天吸烟2次,每次吸烟5支,大约一小时,每周吸烟6天,共吸烟20周。戒烟组每天吸烟2次,每次吸烟10支,共吸烟20周后,脱离吸烟环境10周。采用双抗体夹心酶联免疫吸附法(ABC-ELISA)分别检测血清瘦素、脂联素、IL-6及CRP水平。
     结果
     1.瘦素
     与对照组[(128.00±13.25) pg/ml]比较,大量吸烟组和小量吸烟组及戒烟组[分别为(56.23±8.64) pg/ml; (64.13±10.36) pg/ml; (60.36±7.42) pg/ml]血清水平均降低(P均<0.05);大量吸烟组与小量吸烟组及戒烟组相比,差异均无统计学意义。
     2.脂联素
     与对照组[(0.369±0.032) ng/ml]比较,大量吸烟组、小量吸烟组及戒烟组[分别为(0.322±0.045) ng/ml]; (0.327±0.024) ng/ml; (0.333±0.059) ng/ml]血清水平均降低(P均<0.05);大量吸烟组与小量吸烟组及戒烟组相比,差异均无统计学意义。
     3.IL-6
     与对照组[(32.71±2.86) pg/ml]比较,大量吸烟组、小量吸烟组及戒烟组[分别为(39.67±3.13) pg/ml; (36.58±2.43) pg/ml; (34.27±7.01) pg/ml]血清水平均升高(P均<0.05);大量吸烟组较小量吸烟组及戒烟组水平高(P均<0.05)。
     4.CRP
     与对照组[(494.49±124.44) ng/ml]比较,大量吸烟组、小量吸烟组及戒烟组[分别为(809.50±141.66) ng/ml; (628.91±128.83) ng/ml; (642.60±180.55) ng/ml]血清水平均升高(P均<0.05);大量吸烟组较小量吸烟组及戒烟水平高(P均<0.05)。
     5.相关性分析
     脂联素水平分别与IL-6和CRP呈负相关(分别为r=-0.392,P<0.01;r=-0.631,P<0.05)。
     结论吸烟可使大鼠血清瘦素与脂联素水平下降;IL-6与CRP水平升高,并且随着吸烟量的增加,IL-6及CRP水平进一步增加;戒烟后,瘦素与脂联素水平无明显变化,仍低于对照组,IL-6与CRP水平明显下降,但仍高于对照组;脂联素水平分别与IL-6和CRP呈负相关;以上结果提示瘦素、脂联素等炎症因子参与吸烟所致COPD的全身炎症反应。
Objective:The objective of this paper is to investigate the role of inflammatory factors like serum leptin, adiponectin, interleukin-6 (IL-6) and C-reactive protein (CRP) in the systemic inflammatory response of smoking-induced COPD.
     Methods:32 Male Wistar rats are randomly divided into four groups:group of rats smoking heavily, group of rats smoking fewer, group of rats smoke abatement and a control group, with 8 rats for each group. Rat smoking device was in accordance with Xusanlin smoking device.Except the rats of the control group, all the other rats smoke 2 times one day,6 days one week, for 20 weeks.The first group and the third group smoke 10 cigarettes each time, while the second group 5. The third group of rats quit smoking for 10 weeks after smoking 20 weeks. Serum leptin, adiponectin, IL-6 and CRP levels are measured by ABC-enzyme-linked immunosorbent assay(ELISA).
     Results:
     1. Leptin:Compared to the control group[(128.00±13.25)pg/ml], serum leptin levels of the rats in the other three groups [(56.23±8.64) pg/ml; (64.13±10.36) pg/ml; (60.36±7.42) pg/ml respectively] decrease significantly (all P<0.05); compared to the first group, the difference is not significant in both the second group and the third group (P> 0.05 respectively)
     2. Adiponectin:Compared to the control group[(0.369±0.032) ng/ml], serum adiponectin levels of the rats in the other three groups[(0.322±0.045) ng/ml; (0.327±0.024) ng/ml; (0.333±0.059) ng/ml; respectively] decrease significantly (all P<0.05); compared to the first group, the difference is not significant in both the second group and the third group (P> 0.05 respectively)
     3. IL-6:Compared to the control group, serum IL-6 levels of the rats in the other three groups[(39.67±3.13) pg/ml; (36.58±2.43) pg/ml; (34.27±7.01) pg/ml respectively] increase significantly (all P<0.05); serum IL-6 levels of the rats in the second group and the third group are lower than those of the first group (P<0.05 respectively)
     4. CRP:Compared to the control group, serum CRP levels of the rats in the other three groups [(809.50±141.66) ng/ml; (628.91±128.83) ng/ml; (642.60±180.55) ng/ml respectively] increase significantly (all P<0.05); serum CRP levels of the rats in both second group and the third group are lower than those of the first group (P<0.05 respectively)
     5. Correlation Analysis:Adiponectin levels are inversely related with IL-6 and CRP (r=-0.392, P<0.01; r=-0.631, P<0.05 respectively)
     Conclusions Smoking reduces the levels of serum leptin and adiponectin but increases the levels of serum IL-6 and CRP of rats, further more, serum IL-6 and CRP levels are positively related with smoking status. After smoke quitting, levels of serum leptin and adiponectin are still lower than those fo the control group, while IL-6 and CRP levels decrease, but both of them are higher than those of the control group. What's more, adiponectin level is inversely related with IL-6 and CRP. These results suggest that inflammatory factors such as leptin, adiponectin be involved in the systemic inflammatory response of smoking-induced COPD.
