摘要
从含水量、装载时间、玻璃化时间和种壳4方面探究红水樱桃(Prunus pseudocerasu L. Hongshui)种子玻璃化超低温保存。结果显示,含水量以30%为好;装载以60%PVS_2在4℃下处理20 min为宜;玻璃化以PVS_2室温下处理60 min为宜;种壳决定保存效果,带种壳可获得100%成活率,而去种壳后几乎不萌发,污染严重。种子成活率检测用离体萌发法,经43℃水浴3 min解冻后接种到1/2 MS+1.0 mg/L BA+0.5 mg/L IBA+2.0 mg/L GA+3.0%蔗糖+0.7%琼脂粉培养基上。
